摘要
目的:检测卵巢上皮性癌组织中胰岛素样生长因子-1(IGF-1)和上皮性钙黏连蛋白(E-cad)的表达以及IGF-1作用于卵巢癌SKOV3细胞后E-cad蛋白的表达和细胞侵袭力的改变,探讨IGF-1在上皮间质转化中的作用。方法:采用免疫组化SP法检测30例卵巢上皮性癌、10例卵巢上皮性交界性肿瘤、10例卵巢良性肿瘤及10例正常卵巢组织中IGF-1及E-cad蛋白的表达。以不同质量浓度(0、25、50、100、200μg/L)的IGF-1刺激SKOV3细胞,应用实时荧光定量PCR法及Western blot法检测E-cad mRNA和蛋白的表达,Transwell法检测细胞侵袭力的变化。结果:卵巢上皮性癌、卵巢上皮性交界性肿瘤、卵巢良性肿瘤及正常卵巢组织中IGF-1的阳性表达率分别为73.3%、30.0%、20.0%和10.0%,E-cad的阳性表达率分别为36.7%、60.0%、70.0%和90.0%;与其他3种组织比较,卵巢上皮性癌组织中IGF-1蛋白的阳性表达率升高,E-cad蛋白的阳性表达率降低(P均<0.05);卵巢上皮性癌组织中IGF-1和E-cad的表达均与临床分期、组织学分级、两级分级系统有关(P均<0.05),而与年龄、组织学类型无关(P均>0.05),且在卵巢上皮性癌组织中IGF-1与E-cad蛋白的表达呈负相关(rP=-0.396,P=0.018)。不同质量浓度的IGF-1作用于SKOV3细胞后,E-cad的表达逐渐降低,SKOV3细胞侵袭力增强,且呈剂量依赖性(P<0.05)。结论:IGF-1、E-cad可能与上皮性卵巢癌的上皮间质转化、侵袭转移有关。
Aim: To detect the expressions of IGF-1 and E-cad in epithelial ovarian cancer tissue and the changes of the expression of E-cad and the cell invasion after SKOV3 cells were dealt with IGF-1,so as to study the role of IGF-1 during epithelial-mesenchymal transition( EMT). Methods: Immunohistochemical SP staining was conducted to detect the expressions of IGF-1 and E-cad in tissue from 30 patients with epithelial ovarian cancer,10 patients with borderline ovarian tumor,10 patients with benign epithelial tumor and 10 controls with normal ovary. SKOV3 cells were treated with different concentrations( 0,25,50,100,200 μg/L) of IGF-1,and the expression of E-cad was observed by real-time reverse transcription-PCR and Western blot,and the invasion was detected by Transwell assay. Results: The positive expression rate of IGF-1 in the epithelial ovarian cancer,borderline ovarian tumor,benign epithelial tumor and normal ovarian tissue was73. 3%,30. 0%,20. 0% and 10. 0%,while that of E-cad in the above tissue was 36. 7%,60. 0%,70. 0% and 90. 0%;compared with the other 3 groups,the positive expression rate of IGF-1 was increased and that of E-cad was decreased in epithelial ovarian cancer tissue( P〈0. 05). The expressions of IGF-1 and E-cad were related with the clinical stage,organization classification and two-tier grading system in epithelial ovarian cancer tissue( P〈0. 05),but not with age or pathological type( P〈0. 05). In the epithelial ovarian cancer tissue,IGF-1 and E-cad had negative correlation( rP=- 0. 396,P =0. 018). The expression of E-cad reduced after SKOV3 cells were dealt with IGF-1( P〈0. 05). IGF-1 promoted invasion of SKOV3 cells in a concentration-dependent manner( P〈0. 05). Conclusion: IGF-1 and E-cad may be concerned with EMT and invasion of epithelial ovarian cancer.
