摘要
目的探究雷公藤甲素对乳腺癌细胞MDA-MB-231和MCF-7增殖的抑制作用。方法取培养的乳腺癌细胞MDA-MB-231和MCF-7,用雷公藤甲素按不同浓度处理24 h后提取细胞蛋白和RNA,用Western blot和RT-PCR来检测磷脂酶D1(PLD1)和c-Myc的表达。结果雷公藤甲素能够抑制乳腺癌细胞MDA-MB-231和MCF-7的增殖,并且在MDA-MB-231细胞中下调PLD1和c-Myc的表达。结论雷公藤甲素能够抑制乳腺癌细胞MDA-MB-231及MCF-7的增殖。
Objective To explore the suppression of traditional Chinese medicine triptolide on breast cancer cells. Methods Human breast cancer MDA-MB-231and MCF-7 cells were treated with different concentrations of triptolide for 24 h. Phospholipase D1 (PLD1) and c-Myc proteins were examined by western blot. c-Myc mRNA was examined by RT-PCR. Results Triptolide inhibited the proliferation of both breast cancer cells, and down-regulated the expression of PLD1 and c-Myc in MDA-MB-231 cells. Conclusion Triptolide can suppress the proliferation of MDA-MB-231 and MCF-7 cells.
