摘要
目的了解姜黄素对心房颤动大鼠的心房肌细胞Kir2.1,Kv LQT1,HERG基因表达的影响,并探讨姜黄素治疗房颤的效果及其机制。方法将24只心房颤动大鼠随机分为试验组(12只)和对照组(12只),分别采用姜黄素溶液与生理盐水进行干预。1周后利用酶解法获得大鼠的单个心房肌细胞,采用Real-time PCR及Western Blot技术检测影响内向整流钾电流(I_(K1))的Kir2.1通道、影响缓慢激活延迟整流钾电流(I_(Ks))的KVLQT1通道、影响快速激活延迟整流钾电流(I_(Kr))的HERG通道的mRNA表达情况,内参照基因为GAPDH。结果试验组的Kir2.1通道的mRNA表达水平下调,与对照组比较,差异有统计学意义(t=2.853,P<0.05);试验组的Kv LQT1,HERG通道的mRNA表达水平上调,与对照组比较,差异均有统计学意义(t=1.862,1.758,P<0.05)。结论姜黄素可通过影响心房颤动大鼠的心房肌细胞Kir2.1,Kv LQT1,HERG基因的表达水平,从而发挥对心房颤动的治疗作用。
Objective To learn the effect of curcumin on Kir2. 1, KvLQT1 and HERG gene expression in atrial myocytes of rats with atrial fibrillation, and to investigate the effect and mechanism of curcumin in the treatment of atrial fibrillation. Methods 24 atrial fib-rillation rats were randomly divided into two groups, 12 in curcumin group and 12 in control group. The control group and the curcumin group were intervened by normal saline and curcumin solution respectively. 1 week later, the single atrial myocytes of rats were obtained by enzymatic method. Real time PCR and Western blot techniques were used to detect the mRNA expression of Kir2. 1 channel which affected inward rectifier potassium current ( IK1 ) , KVLQT1 channel which affected slow activation of delayed rectifier potassium current ( IKs ) , and HERG channel which affected fast activation of delayed rectifier potassium current ( IKr ); and internal reference gene was GAPDH. Results The expression level of mRNA of Kir2. 1 channel in curcumin group was down regulated, and the difference was sta-tistically significant ( t=2. 853, P 〈 0. 05 ) in comparison with control group;The mRNA expression level of KvLQT1 and HERG chan-nels in curcumin group were up regulated, and the difference was statistically significant ( t=1. 862, 1. 758, P 〈 0. 05 ) in comparison with control group. Conclusion The effect of curcumin on atrial fibrillation was induced by Kir2. 1, KvLQT1 and HERG gene expres-sion in atrial myocytes of rats with atrial fibrillation.
