熊去氧胆酸对AFP-L3及GGTmRNA-H亚型表达的影响 被引量:2

摘要 [目的]探讨熊去氧胆酸(UDCA)对甲胎蛋白异质体(AFP-L3)及大鼠肝组织与血液中谷氨酰转移酶mRNA-H(GGTmRNA-H)亚型表达的影响,进一步研究UDCA对肝癌发生的影响。[方法]Wister大鼠92只随机分成5组:模型组(24只)、干预A组(24只)、干预B组(24只)、正常对照组(10只)、UDCA对照组(10只)。对模型组、干预A组、干预B组以二乙基亚硝铵腹腔注射诱导肝癌形成后,干预A组给予含2.5g/L的UDCA饮用水,干预B组给予含5g/L UDCA的饮用水;正常对照组给予0.9%氯化钠溶液腹腔注射,UDCA对照组给予含5g/L UDCA的饮用水。通过ELISA法检测各组大鼠血清中AFP-L3的水平,采用实时定量PT-PCR检测GGTmRNAH亚型在各组大鼠血液及肝组织的表达情况。[结果]1模型组、干预A组、干预B组AFP-L3绝对值明显高于正常对照组、UDCA对照组(P<0.05)。干预A组、干预B组AFP-L3水平均低于模型组(P<0.05)。干预A组、干预B组间比较差异无统计学意义(P>0.05)。2干预A组、干预B组中GGTmRNA-H亚型在癌组织及癌旁组织的表达量均较模型组明显下降(P<0.05);干预A组、干预B组血液中的GGTmRNA-H亚型也较模型组下降(P<0.05);但干预A组、干预B组间比较差异无统计学意义(P>0.05)。[结论]1UDCA能降低大鼠肝癌发生过程中AFP-L3的水平,提示UDCA对肝癌的发生有一定的预防作用;2UDCA能降低肝癌大鼠肝癌组织及血液中GGTmRNA-H亚型的表达,二者间接反映UDCA能降低肝癌的发生,对肝癌的发生有一定预防作用。 [Objective]To explore the effect of ursodeoxycholic acid(UDCA)on AFP-L3 and r-glutamine shift enzyme(GGT)H subtypes gene expression in rat hepatoma model. Furtherly,to investigate the influ- ence of UDCA on the happening of liver cancer. [Methods]A total of 92 Wistar rats were randomly divided into 5 groups.model group only, UDCA intervention group A, UDCA intervention group B, the normal con- trol group and UDCA control group. DEN solutions were injected intraperitoneally and different concentra- tions of UDCA were fed in intervention group A(2.5 g/L) ,B(5 g/L). Blood was drew from the heart every four weeks from the tenth week after the DEN solutions were injected and the serum AFP-L3 was detected by ELISA. The expression of GGTmRNA-H subtypes in the blood and liver tissue in all the rats were de- tected by the real-time quantitative(PT-PCR). [Results] ①In model group and UDCA intervention A, B groups,the levels of AFP-L3 were significantly higher than that of normal control group and UDCA con- trol group(P〈0.05). In UDCA intervention group A and B, AFP-L3 levels were lower than the model group(P〈0.05). There were no significant differences betweenUDCA intervention grout) A and B(P〈0.05). ②In UDCA intervention group A and B,the expression of GGTmRNA-H subtypes in cancerous tis- sue and adjacent noncancerous tissue was significantly lower than those in model group(P〈0.05). Similar changed happened in the blood samples. [Conclusion]①UDCA can reduce AFP-L3 level in rat liver cancer process,which suggests that UDCA has certain preventive effect in liver cancer happening. ②UDCA can reduce expression of GGTmRNA-H subtypes in both liver cancer tissue and blood,which indirectly reflects UDCA can reduce the occurrence of liver cancer and have certain preventive effect to liver tumors.
