微生物源α-L-鼠李糖苷酶制备橙皮素单葡萄糖苷 被引量:2

摘要 橙皮苷是一种黄酮类化合物,具有抗炎、抗氧化、抗癌、维持血管正常渗透压和降低血脂等多种生理活性。文献报道大多数天然产物中由于鼠李糖糖苷的存在,会降低了其生物利用率。从腐烂的柑橘中筛选得到一株α-L-鼠李糖苷酶的产生菌株,能将橙皮苷水解得到水溶性更好的橙皮素单葡萄糖苷。进一步对α-L-鼠李糖苷酶进行了分离纯化及酶学性质的研究。最后,采用大孔树脂法对橙皮素单葡萄糖苷进行了分离纯化,并对其洗脱工艺进行了研究,最后确定的工艺为30%的乙醇水溶液在1mL·min-1的流速下,可以有效的分离得到橙皮素单葡萄糖苷,其纯度达到87.55%。 Hesperidin is a type of flavonoid compound,with great activities of anti-inflammatory effects,antioxidant,anticancer,maintenance of vascular normal osmotic pressure,reducing blood lipids and other physiological activities.The existence of rhamnose glycosidic in most natural products would reduce their bioavailability.In this study,we screened aα-L-rhamnosidase-producing strain from rotten citrus that could hydrolyze hesperetin to get a better water-soluble copound Hesperetin monoglucoside(HMG).The purification and characterization of alpha-L-rhamnosidase were also studied.Finally,a macroporous resin method was used to isolate and purify Hesperetin monoglucoside.Our results revealed that using 30%aqueous ethanol with a flow rate of 1 mL· min-1 can effectively isolate Hesperetin monoglucoside,and its purity reached 87.55%.
