响应面法优化SFE-CO_2提取酸枣仁中脂肪油及成分分析 被引量:4

Supercritical CO_2 Fluid Extraction of the Fatty Oil FromZizyphi spinosi Semen Optimized by Response Surface Methodology

摘要 以酸枣仁为研究对象,采用超临界流体萃取技术提取油酸枣仁中脂肪油成分,并利用响应面分析法分析了超临界流体萃取的试验条件,研究了系统压力、提取温度、动态萃取时间和酸枣仁粉末粒度4个影响因素对脂肪油提取效果的影响,并应用GC-MS技术对脂肪油中的主要成分进行了分析。结果表明:应用响应面法优化酸枣仁中脂肪油的SFE-CO_2最佳提取方案为提取温度40℃,萃取压力395bar,动态萃取时间2.5h,酸枣仁样品粉末粒度为40目,其中脂肪油提取效果较好,提取率约为20%;应用GC-MS检测结果表明,脂肪油中的主要成分为10-十八碳烯酸、十八烷酸、9,12-十八碳二烯酸、十六烷酸等;脂肪油是其主要成分,占相对含量的60%以上;SFE-CO_2萃取技术具有高效、方便、提取效率高、绿色环保等显著优点,适合于酸枣仁中脂肪油的提取。 Taking Zizyphi spinosi Semen as material,supercritical CO_2 fluid extraction of the fatty oil from Zizyphi spinosi Semen was carried out,the extraction temperature,pressure,time and the sample particle size were independent determined and the conditions were evaluated by response surface methodology(RSM).The results showed that the maximum extraction yield of about 20% was achieved at extract temperature of 40 ℃,pressure of 395 bar,dynamic extraction time of 2.5hand power particle size of 40 mesh.The chemical compositions of fatty oil were investigated by GC-MS.The results indicated that supercritical CO_2 fluid extraction method was the suitable method,the main fatty acids were 10-octadecenoic acid,octadecanoic acid,9,12-octadecadienoic acid and palmitic acid,respectively.Supercritical fluid extraction was proved to an effective technique for extracting fatty oil fromZizyphi spinosi Semen.
