沉默H19对骨肉瘤细胞系MG63增殖、侵袭的影响 被引量:3

摘要 目的:研究长链非编码RNA H19对骨肉瘤MG63细胞增殖、侵袭的影响。方法:RNA干涉技术沉默MG63细胞中H19,荧光实时定量PCR(RT-q PCR)检测骨肉瘤MG63细胞中H19的表达情况;流式细胞术检测细胞周期的变化;MTT实验检测沉默H19表达对细胞增殖的影响;Transwell侵袭实验检测细胞侵袭能力的变化。结果:H19-siRNA可有效沉默H19在骨肉瘤细胞MG63中的表达,沉默MG63细胞中H19的表达可以明显抑制骨肉瘤细胞的细胞周期,降低骨肉瘤细胞的增殖、侵袭能力。结论:H19在骨肉瘤的发生发展及侵袭中发挥重要作用。 Objective: To investigate the effects of lncRNA H19 on the proliferation and invasion of human osteosarcoma MG63 cells. Methods: MG63 cells were transfected with siRNA or control siRNA for 48 hours,and then detect the H19 mRNA expression by quantitative Real- time PCR( RT- q PCR). After siRNA and control siRNA transfected into MG63 cells for 48 hours,the proliferation of MG63 cells were analyzed by MTT,the cell cycle of MG63 cells was analyzed by Flow cytometry,the MG63 cells invasion ability was analyzed by Transwell cell invasion assay.Results: The mRNA expression level of H19 in MG63 cells was effectively silenced by H19- siRNA and MG63 cells proliferation and cell cycle was inhibited after transfected lncRNA H19- siRNA,the invasion ability of MG63 cells decreased after transfected H19- siRNA. Conclusion: H19 may play an important role in the progression,invasion of human osteosarcoma.
