摘要
目的:研究CGMCC No.8730冠突散囊菌标准化发酵的金花黑茶水提物及全茶粉对排便以及肠道菌群的影响。方法:雄性成年BALB/c小鼠160只,随机分为正常对照、阴性对照、阳性对照、低、中、高剂量,毛茶水提物、全茶粉8组。相应试剂灌胃14 d后无菌取新鲜粪便进行肠道菌群16S r DNA测序,随后盐酸洛哌丁胺造小肠蠕动抑制便秘模型,测定各组小肠推进率、首粒黑便时间、排便粒数与排便量。结果:(1)3种茶制剂均能显著增强洛哌丁胺造成的小肠蠕动抑制模型小鼠的小肠运动(P<0.05);(2)250、750 mg/kg·bw剂量水提物和全茶粉组首粒黑便时间显著低于阴性对照组(P<0.05),粪便粒数和粪便质量显著高于阴性对照组(P<0.05);(3)与阴性对照组相比,金花黑茶全粉组双歧杆菌、乳球菌百分比增幅较大,大肠杆菌增幅较小,同时ACE指数、Shannon指数增加。结论:CGMCC No.8730冠突散囊菌标准化发酵的金花黑茶水提物和全茶粉在BALB/c小鼠中具有增加小肠运动、缩短排便时间、增加排便量的作用,其全茶粉具有调节肠道菌群的作用。
Objective: To investigate the effect of the CGMCC No.8730 Eurotium cristatum fermented dark tea on defecation and gut microbiota. Methods: 160 male BALB/c mice were randominze into 8 groups, comprising normal control, negative control and positive control, low, medium and high dose, row tea water extract and tea powder. All by intraperitoneal injection for 14 days, small intestine propulsion rate, time of first black faeces, count and volume of faeces were tested in loperamide induced constipation model, fresh faeces was collected for 16 S r DNA sequencing. Results:(1)250 mg/kg·bw and and above, as well as other two kinds of tea preparation could enhance small intestine movement(p〈0.05), in which Eurotium cristatum fermented dark tea water extract showed the strongest effect.(2)Time of first black faeces was significantly lower in 250 mg/kg·bw, 750 mg/kg·bw dose groups and tea powder group than that of negative control group(p〈0.05), and count and volume of faecal in 250 mg/kg·bw, 750 mg/kg·bw dose groups were significantly higher(p〈0.05). The effect of Eurotium cristatum fermented dark tea water extract on first black faeces, count and volume of faeces showed a dose-response effect.(3)Compared with control group, in Eurotium cristatum fermented dark tea podwer group the percentage of Bifidobacterium and Lactococcus showed larger increment, while the increase of Escherichia was smaller, the abundance and diversity of gut microbiota rised. Conclusion: Both CGMCC No.8730 Eurotium cristatum fermented dark tea water extract and powder could enhance small intestine movement, shorten defecation time, increase the amount of defecation in BALb/c mice, the powder could improve gut microbiota.
