海红果酒多糖分离纯化及结构初步分析

Preparation and structure characterization of polysaccharide from Malus micromalus Makino wine

摘要 海红果酒经过超滤、AB-8大孔树脂吸附,DEAE-52纤维素柱分离得到MWPS-Ⅰ、MWPS-Ⅱ、MWPS-Ⅲ3种多糖。通过Sephadex-20葡聚糖凝胶层析柱和透析对多糖进一步纯化,采用毛细管电泳、紫外光谱和红外光谱对海红果酒多糖MWPS-Ⅱ的结构进行初步研究。结果表明:多糖MWPS-Ⅱ由鼠李糖、甘露糖、阿拉伯糖、葡萄糖及葡萄糖醛酸5种单糖组成。UV检测显示海红果酒多糖在(260-280)nm处未见核酸和蛋白质等生物大分子的吸收峰,经红外检测证实MWPS-Ⅱ具有多糖的特征吸收峰。 A water-soluble polysaccharide namely MWPS-Ⅱ from Malus micromalus Makino wine was obtained by successive purification using Macro porous resin AB-8, cellulose DEAE-52 and Sephadex LH-20 gel-filtration chromatography. The structural characterization of the purified polysaccharide fraction was evaluated in this paper. The result showed that MWPS-Ⅱ was a homogeneous heteropolysaccharide including rhamnose, mannose, glucose, arabinose and glucuronic acid. MWPS-Ⅱ has a significant absorption peak in the wavelength of 200 nm. This is the typical absorption of polysaccharides. There was not be detected significant absorption peak in the wavelength of 280 nm and 260 nm by an ultraviolet spectrophotometer, which indicates that protein and nucleic acid impurity have been reduced to the lowest amount. Characterization of MWPS-Ⅱ by FT-IR analysis showed the typical absorption of polysaccharide.
