摘要
目的观察甲基乙二醛(MG)对海马神经元凋亡相关蛋白表达的影响及远志皂苷元对其的干预作用。方法将体外培养的海马神经元,分为对照组、模型组及远志皂苷元低、中、高剂量组。模型组用MG处理,远志皂苷元低、中、高剂量组分别应用不同剂量的远志皂苷元(1.0、2.0、4.0 g/m L)预孵育12 h,再与MG共培养24 h。采用MTT法检测细胞存活率,Western blot法检测MG对海马神经元凋亡相关蛋白Caspase-3、Bax和Bcl-2表达的影响,以及远志皂苷元的干预效果。结果远志皂苷元能明显改善海马神经元存活率;与对照组相比,模型组Caspase-3和Bax表达明显升高,Bcl-2表达降低;与模型组相比,远志皂苷元各剂量组Caspase-3和Bax表达明显降低,Bcl-2表达升高。结论远志皂苷元在一定程度上可抑制MG引起的细胞凋亡,保护海马神经元。
Objective To investigate the effects of methylglyoxal (MG) on the expression of related proteins in apoptosis of hippocampal neurons and intervention of tenuigenin. Methods Hippocampal neurons were cultured in vitro, and were divided into the control group, model group and low-dose, moderate-dose and high-dose tenuigenin groups with tenuigenin ( 1.0 , 2. 0, and 4. 0 g/ mL) pre-incubated for 12h respectively, and co-cultured with MG for 24 h. Cell survival rates were assessed by MTI'. The expressions of Caspase-3, Bax and Bcl-2 related with cell apoptosis of hippocampal neurons, as well as the effect of intervention of tenuigenin were detected by Western blot. Results Cell survival rates could be improved significantly by tenuigenin. The expression of Caspase-3 and Bax were increased, Bcl-2 protein decreased compared with the control group; while the expressions of Caspase-3 and Bax were decreased and Bcl-2 protein increased compared with the model group. Conclusion Tenuigenin could inhibit cell apoptosis caused by MG in a certain degree and protect hippocampal neurons.
