苦酸通调方对自发性糖尿病大鼠腹部脂肪组织中SCAP及SREBP-1c蛋白表达的影响

摘要 目的观察苦酸通调方对自发性糖尿病大鼠腹部脂肪中激活蛋白(SCAP)及固醇调节元件结合蛋白-1c(SREBP-1c)蛋白表达的影响。方法将高脂饲料喂养的雄性SPF级ZDF大鼠随机分为模型组、苦酸通调组、吡格列酮组,普通饲料喂养的雄性SPF级ZL大鼠为正常对照组,每组10只。苦酸通调组按照3.29 g/kg/d灌胃给药,吡格列酮组按照1.07 mg/kg/d灌胃给药,连续给药12周。经Western blot法检测各组大鼠腹部脂肪组织中SCAP及SREBP-1c蛋白的表达。结果与正常对照组比较,模型组SCAP及SREBP-1c蛋白表达明显升高(P<0.05)。与模型组比较,苦酸通调方组及吡格列酮组可明显降低SCAP及SREBP-1c蛋白表达(P<0.01)。结论苦酸通调方可明显降低糖尿病大鼠腹部脂肪组织中SCAP及SREBP-1c蛋白的表达。 Objective To investigate the effect of Kusuan Tongtiao Formula on the expression of SCAP and SREBP-1c protein in abdominal adipose tissue of spontaneously diabetic rats. Methods Male SPF level ZDF rats( Zucker Diabetic Fatty rats) induced by feeding of high-fat and high-carbohydrate diet were randomly divided into the model group( MG),Kusuan Tongtiao group( KG) and Pioglitazone group( PG). A normal control group( NG) was also set up by male Zucker Lean( ZL) rats which were fed with normal diet. There were 10 rats in each group. Kusuan Tongtiao group and Pioglyitazone group were given Kusuan Tongtiao Decoction3. 29 g / kg / d and Pioglitazone suspension 1. 07 mg / kg / d by gavage respectively for twelve weeks. The expression of SCAP and SREBP-1c protein in abdominal adipose tissue of spontaneously diabetic rats were detected by Western blot method. Results The expression of SCAP and SREBP-1c protein in abdominal adipose tissue in model group were significantly higher than that in normal control group( P 0. 05). Compared with MG,levels of the expression of SCAP and SREBP-1c protein in Kusuan Tongtiao group and Pioglyitazone group were decreased obviously( P 0. 01). Conclusion Kusuan Tongtiao Formula can inhibit the expression of SCAP and SREBP-1c protein in abdominal adipose tissue of diabetic rats.
