摘要
目的:观察祛痰通络汤对糖尿病大鼠肾组织中内质网应激相关分子GRP78和CHOP表达的影响。方法:高糖高脂饲料联合腹腔注射STZ制备糖尿病模型,随机分为4组,分别为正常对照组、模型组、祛痰通络汤组、四苯基丁酸组,治疗8周。观察各组24 h尿蛋白定量、肾脏病理学变化;免疫组化和western-blot检测肾组织内质网应激相关蛋白GRP78,CHOP的表达。结果:祛痰通络汤降低尿蛋白,减轻病理改变;模型组肾组织的GRP78、CHOP表达明显升高(P<0.05),祛痰通络汤组GRP78、CHOP表达明显低于模型组(P<0.05)。结论:糖尿病大鼠肾脏中GRP78、CHOP表达明显增强,祛痰通络汤降低内质网应激,可能是其肾脏保护作用的重要机制。
Objective: To investigate the effects of Qutantongluo Decoction on the expression of Endoplasmic reticulum stress- related molecules GRP78 and CHOP in renal cortex of diabetic rats. Methods: The diabetes rat model was induced by feeding high- glucose high- fat diet in combination with intraperitoneal injection of STZ,Wistar rats were randomly divided into 4 groups: normal control group、the model group、Qutantongluo Decoction group、4- phenyl butyric acid treatment( PBA group). After 8 weeks,24 hour urinary protein excretion were determined,renal tissue were were analyzed by pathological examination,the expression of glucose- regulated protein 78( GRP78) 、C / EBP homologous protein( CHOP) were examined by immunohistochemistry and western- blotting respectively. Results: Qutantongluo Decoction treatment significantly reduced proteinuria,improved pathological status,the expressions of GRP78 and CHOP in model group was significantly higher than control group; Qutan Tongluo Decoction reduced the expressions of GRP78( P〈0. 05) and CHOP( P〈0. 05) compared with the model group. Conclusion: Endoplasmic reticulum stress- related molecules GRP78 and CHOP was abundantly expressed in the kidneys of diabetic rats,Qutan Tongluo Decoction decreased ER stress,which might be an important mechanism in the renoprotective functionin of DN.
