摘要
人源性长寿保障基因2(LASS2)是中国发现并命名的一个肿瘤转移抑制基因,也被称作肿瘤转移抑制相关基因1(TMSG-1),能够通过抑制V-ATPase质子泵活性、调节神经酰胺表达、调控转录过程等途径抑制人类多个系统的肿瘤的生长、浸润及转移,LASS2的表达与肿瘤的临床分期、预后和耐药性密切相关,同时有新的研究提出LASS2/TMSG-1能够参与调控正常细胞的迁移从而促进神经损伤的修复,现就LASS2/TMSG-1基因在相关疾病中表达的意义及可能的作用机制和应用前景的研究现状做一综述。
The homosapiens longevity assurance homologue 2(LASS2),which is also called tumor metastasis suppressor gene-1(TMSG-1),is a newly identified tumor suppressor gene.Increasing amount of studies have showed that LASS2/TMSG-1plays important roles in promoting apoptosis,inhibiting invasion and metastasis of tumor cells,the molecular mechanisms of LASS2/TMSG-1 suppressing cancer remain uncertain at present,several studies have pointed out the inhibitory effect may be related to interaction with vacuolar H+-ATPases and ceramid expression regulating,in addition,LASS2/TMSG-1 is possibly involved in transcriptional regulation.It's suppressive function is also a key regulator in cell migration and may highly correlated to recurrence and prognosis in related human diseases,these findings provide an important insight of noval therapeutic target in gene therapy.
