摘要 Edmonton方案的提出推动了胰岛移植的加速发展.雷帕霉素作为Edmonton方案中推荐的免疫抑制药物得到了人们更多的关注.雷帕霉素的衍生物(依维莫司、地磷莫司、佐他莫司、替西罗莫司)也引起了人们的兴趣.人们在探讨他们的免疫抑制作用和抗肿瘤作用的同时,发现他们可引起血糖升高等胰岛毒性症状.他们能否成为合适的胰岛移植抗排斥药物还有待进一步研究.本文就雷帕霉素及其衍生物的作用、应用进展和他们对胰岛的毒性作用作一综述. The development of islet transplantation has been promoted by the proposal of the Edmonton protocol. Rapamycin, as a recommended immunosuppressive medicine of the Edmonton protocol, has been getting extraordinarily popular. At the same time, derivatives of rapamycin (everolimus, deforolimus, zotarolimus and temsirolimus) have also garnered great interest. While the immunosuppressive and anti-cancer effects of rapalogs were being discussed actively, researchers discovered their cytotoxic effect on pancreatic islets. Whether they could be ideal drugs for anti-rejection after islet transplantation needs further study. This review aims to elucidate the function and application of rapalogs as well as their toxicity to pancreatic islets.
