摘要
目的:分析中国不同人群中葡萄糖转运蛋白4基因多态性分布特征,寻找与运动能力关联的多态位点,作为中长距离游泳项目选材用基因标记。方法:选择120名中国优秀游泳运动员,采用飞行时间质谱(TOF-MS)解析葡萄糖转运蛋白4(GLUT4)rs5418位点;对照组为206名普通大学在校学生,多态位点的分析采用常规方法 PCR-RFLP。结果:1)运动员组和对照组的基因型分布频率都符合Hardy-Weinbery平衡;2)实验组rs5418的基因型分布频率与对照组相比有显著性差异(χ^2=6.338,df=2,P=0.042),携带AA纯合子型的游泳运动员显著性高于对照组;3)rs5418等位基因分布,2组具有显著性差异(χ^2=6.171,df=1,P=0.008),对照组中A型等位基因极其低于运动员组;4)AA纯合型与AG+GG复合基因型分布,实验组和对照组有显著性差异(χ^2=5.877;df=1,P=0.015)。结论:GLUT4基因rs5418多态位点与优秀中长距离游泳运动能力关联,可以作为该项目选材用分子标记。
The purpose of the study was to investigate the association between the GLUT4 polymorphism and elite middle-long distance( MLD) swimmers' performance. Methods: The GLUT4 rs5418 of 120 elite MLD swimmers was tested by TOF-MS,and other 206 normal college students' rs5418 polymorphism was measured by PCRRFLP. Results: 1) Genotype distributions of both athletes and normal students were in Hardy-Weinberg equilibrium. 2) There was difference of GLUT4 rs5418 polymorphism between elite swimmers and normal students,AA genotype of athletes was significant higher than that of students( χ^2= 6. 338,df = 2,P = 0. 042). 3) There was difference of rs5418 allele between elite swimmers and normal students( χ^2= 6. 171,df = 1,P = 0. 008),A allele of normal students was much lower than that of athletes. 4) There were differences in AA genotype and AG +GG genotype between elite swimmers and normal students( χ^2= 5. 887,df = 1,P = 0. 015). Conclusion:GLUT4 rs5418 polymorphism is associated with Chinese elite MLD swimmers' performance,and it can be one of the genetic marks for Chinese elite MLD swimmers selection.
