Relationship between Girdin Expression and Invasion and Metastasis of Non Small Cell Lung Cancer and Its Prognosis

摘要 目的:探讨肌动蛋白结合蛋白Girdin与非小细胞肺癌(Non small cell lung cancer,NSCLC)侵袭转移的关系及其对预后的影响。方法:应用免疫组织化学法检测167例非小细胞肺癌患者的病理组织标本中Girdin蛋白和MMP-9的表达。结果:在167例组织标本中Girdin蛋白高表达率为38.9%,其表达水平与患者分期、淋巴结转移、远处转移及生存状况密切相关,差异有统计学意义(P<0.05),而与患者性别、年龄、吸烟指数、评分、病理类型及分化程度均无关(P均>0.05)。Girdin蛋白的高表达往往伴有MMP-9的高表达,两者显著相关,且差异有统计学意义(P<0.05)。Kaplan-Meier单因素生存分析显示Girdin高表达为非小细胞肺癌患者预后的不良因素(P<0.05)。COX多因素回归分析显示Girdin的表达水平和分期是判断预后的独立指标(P<0.05)。结论:Girdin蛋白在非小细胞肺癌的侵袭和转移中可能发挥着重要的作用,在判断非小细胞肺癌患者预后方面具有一定的价值。 Objective: To explore the correlation between Girdin and invasion and metastasis of NSCLC, and the impact of Girdin on prognosis. Methods: Immunohistochemistry was used to detect the expression of Girdin and MMP-9 in 167 tissue samples of Non small cell lung cancer. Results: High expression of Girdin protein was found in 38.9%(65/167) of tissue samples and was significantly associated with stage, lymph nodes metastasis, distant metastases and survival(P〈0.05). No correlation were found between expression of Girdin and gender, age, smoking status, PS, pathological type and differentiation(P〈0.05). High expression of Girdin was found to be correlated with high expression of MMP-9(P〈0.05). Kaplan-Meier survival analysis showed high expression of Girdin was a poor prognostic factor in NSCLC(P〈0.05). The multivariate Cox model identified expression of Girdin and stage as independent prognostic factors. Conclusions: Girdin plays an important role in invasion and metastasis of NSCLC, and it is useful for evaluating the prognosis of NSCLC.
