夹脊电针对ALS-SOD1^(G93A)转基因小鼠病程进展和行为学的影响 被引量:8

Effect of Electroacupuncture at Jiaji point( EX- B2) on Disease Progression and Behavior in the Transgenic Mouse Model of Amyotrophic Lateral Sclerosis

摘要 目的:通过夹脊电针干预ALS-SOD1^(G93A)转基因小鼠,观察其对小鼠病程进展和行为学的影响。方法:本实验以ALS-SOD1^(G93A)转基因小鼠为实验对象,应用美国Jackson实验室提供的引物和方案对ALS-SOD1G^(G93A)转基因小鼠进行PCR鉴定。根据病情进展观察小鼠的表型特征。30只ALS-SOD1^(G93A)转基因小鼠,随机分为电针组、手针组和模型组,每组10只(雌性7只,雄性3只),分别给予电针、手针、抓握及捆绑处理,联合应用悬尾试验、称重、转棒实验来观察小鼠的发病时间和生存期。结果:与模型组相比,电针组发病推迟了8天,有显著性差异(P=0.023),手针组发病推迟了5天,但无显著性差异(P>0.05)。与模型组相比,手针组、电针组小鼠生存期分别延长了5天和10天,有显著性差异(分别为P=0.039和P=0.000),与手针组比较,电针组生存期延长了5天,有显著性差异(P=0.042)。与模型组相比,电针治疗能够明显延缓小鼠体重丢失,有显著差异(P<0.05),手针组小鼠体重丢失亦得到一定的延缓,但没有看到显著作用(P>0.05)。与模型组相比,手针组和电针组转棒运动功能明显得到改善,分别延长了1周和2周。结论:夹脊电针能够延迟ALS-SOD1^(G93A)转基因小鼠的发病,延长小鼠的生存期,改善小鼠的运动功能障碍。 Objective: This study was aimed to observe the effect of electroacupuncture at Jiaji point( EX-B2) on disease progression and behavior in the transgenic mouse model of ALS( ALS- SOD1^(G93A)). Methods:ALS- SOD1^(G93A)transgenic mice were used for this study. We identified the ALS- SOD1^(G93A)transgenic mice by PCR with the primer sequence and identification methods provided by Jackson Laboratory,and observed the phenotype of ALS- SOD1^(G93A)transgenic mice. 30 ALS- SOD1^(G93A)transgenic mice were divided into an electroacupuncture group( group A),an acupuncture group( group B) and a control group( group C),with 10 in each group. Each group had 7 females and 3 males. ALS- SOD1^(G93A)transgenic mice were treated by electroacupuncture,acupuncture,seizing and binding. Disease progression of each group was monitored 2 times / week,including suspending tail test,weight and rotarod test. Results: In this study,group A delayed the onset of the disease by 8 days compared with group C,and the difference was statistically significant( P = 0. 023). Group B also slightly increased this value compared with group C,although there was no statistical significance( P 0. 05). Group A and group B respectively prolonged the survival time by 10 days and 7 days compared with group C,and the differences were statistically significant( P = 0. 039,P = 0. 000),and group A prolonged the survival time by 5 days compared with group C,and the differences were statistically significant( P = 0. 042).Group A delayed the loss of mice weight compared with group C,and the difference was statistically significant( P 0. 05); though group B also delayed the loss of weight,there was no significant difference( P 0. 05).Group A and group B could significantly improve performance of rotarod test compared with group C,and the difference was statistically significant. Conclusion: This study shows that electroacupuncture at Jiaji point( EX- B2) may improve motor function,delay the onset of disease and prolong the survival time.
