摘要
目的探讨氟伐他汀对同型半胱氨酸(Hcy)诱导的人脐静脉内皮细胞(HUVECs)凝集素样氧化低密度脂蛋白受体-1(LOX-1)信使核糖核酸(m RNA)表达的影响。方法分离培养HUVECs,分为正常对照组、Hcy刺激组:100μmol/L Hcy与细胞孵育24 h,3种不同浓度的氟伐他汀干预组:分别以不同浓度的氟伐他汀(0.1、1、10μmol/L)预孵育细胞24 h,再与100μmol/L Hcy共同培养24 h,应用逆转录-聚合酶链反应(RT-PCR)检测LOX-1 m RNA的表达。结果 Hcy显著诱导HUVECs LOX-1 m RNA的表达,氟伐他汀抑制Hcy诱导的LOX-1 m RNA的表达,并呈现一定剂量依赖性。结论 Hcy可能通过LOX-1介导其内皮损伤作用,氟伐他汀可能通过减少LOX-1过度表达发挥其抗动脉粥样硬化作用。
Objective To investigate the effect of fluvastatin on the expression of LOX-1 induced by homocysteine (Hcy) in cultured human umbilical vein endothelial cells (HUVECs). Methods Human umbilical vein endothelial ceils were cultured and treated with Hcy (100μmol/L for 24 hours) alone, or pre-treated with fluvastatin (0.1, 1 or 10 μmol/L for 24 h), followed by Hcy treatment. After respective treatment, the expression of lectin-like ox-LDL receptor-1 (LOX-1) was measured by reverse transcription-polymerase chain reaction (RT-PCR). Results Hcy (100 umol/L) treatment up regulated the expression of LOX-1 in HUVECs. Pretreatment of HUVECs with fluvastatin (0.1, 1 or 10 μmol/L) de- crease Hcy-induced expression of LOX-1. A high concentration of fluvastatin (10 umol/L) was more potent than the lower concentration (1 μmol/L). Conclusion These observations indicate that endothelial dysfunction elicited by Hcy- may beassociated with an increase of LOX-1 expression. Inhibition of expression of LOX-1 may be another mechanism of beneficial effects of statins in vascular diseases.
