摘要
目的 探讨鼠诺如病毒(Murinenoroviurs,MNV)感染C57BL/6小鼠后临床及生物学特征,了解MNV的感染特点。方法将MNV—CWI毒株以高剂量(10^4PFU/只)和低剂量(102PFU/只)分别灌胃到C57BL/6小鼠体内,建立MNV感染小鼠模型。对感染小鼠进行体征观察和体温检测,检测粪便和血液排毒情况、组织中病毒的分布情况以及十二指肠病理切片检查。结果感染的小鼠没有明显的体表变化,没有腹泻、发热等临床症状和病理改变。粪便检测感染后1~2d排毒。在感染后第3天部分血液中检测到MNVRNA。主要感染部位为消化系统和脾脏。结论本研究建立的MNV—CW1感染C57BL/6小鼠模型为MNV感染小鼠致病机制进一步研究提供实验信息和技术支撑。
Objective To establish the animal model of infection of C57BL/6 mice by a murine norovirus (MNV) strain MNV-CW1. Methods C57BL/6 mice were orally administrated with MNV-CW1 of 104 PFU or 10z PFU per mouse. After establishment of mice model, we observed the clinical signs of disease and detected the virus RNA in feces, serum and organs. Results MNV-CW1 cannot induce obvious clinical syndrome in C57BL/6 mice, such as fever, diarrhea and histophathological changes. MNV RNA was detected in fecal on day 1 or 2 after infection and the main infection sites were spleen and digestive system of mice. Conclusions An animal model of MNV-CW1 was established and it will provide a basis for the further study of molecular pathogenesis of MNV infection.
