摘要
目的采用高效液相色谱法,建立测定枸杞根中绿原酸和阿魏酸含量的方法。方法色谱条件:Boston C18柱(4.6 mm×250 mm,5μm);流动相:乙腈-2%冰醋酸水溶液梯度洗脱;流速:1 m L/min;柱温:30℃;检测波长:324 nm。结果绿原酸和阿魏酸分别在进样量20.54~92.43 ng和4.10~18.38 ng范围内线性关系良好,平均回收率(n=6)分别为98.2%和100.9%,RSD分别为1.44%和1.85%。结论本法准确可靠,重现性好,可用于测定枸杞根中绿原酸和阿魏酸的含量。
OBJECTIVE To establish an HPLC method for the determination of chlorogenic acid and ferulic acid in gouqigen.METHODS The chromatographic separation was performed on Boston C18column( 4. 6 mm × 250 mm,5 μm) with acetonitrile-2%acetic acid aqueous solution gradient elution. The flow rate was 1 m L / min at 30 ℃,and the detection wavelength was set at 324 nm.RESULTS The linearity for chlorogenic acid and ferulic acid in gouqigen were in the range of 20.54-92.43 ng( r = 0.999 8,n = 6)and 4.10- 18. 38 ng( r = 0. 999 9,n = 6),respectively. The average recovery were 98. 2%( RSD = 1. 44%) for chlorogenic acid and100.9%( RSD = 1.85%) for ferulic acid. CONCLUSION The method is accurate,reliable and reproducible,it could be used as a quality control method for chlorogenic acid and ferulic acid in gouqigen.
