摘要
目的:探讨连续跳跃运动条件下兔髌骨髌腱结合部内外侧区域形态学,以及胶原蛋白、血管生成等相关生长因子的表达特征。方法:18周龄雌性新西兰大白兔10只,随机分为跳跃组(JE)和对照组(Con),每组5只,JE组每天在电刺激下跳跃150次,每周训练5天,对照组不予任何处理。4周后,取双下肢髌骨髌腱复合体,行HE染色、番红染色和免疫组化染色。结果:与Con组相比,JE组髌骨髌腱结合部细胞密度、纤维软骨带厚度、粘多糖蛋白区域面积显著增加,JE组髌骨髌腱结合部内侧区域细胞密度显著低于外侧区域(P<0.05),内侧区域纤维软骨带厚度显著高于外侧区域(P<0.05);与Con组相比,JE组髌骨髌腱结合部CollagenⅢ,HIF-1α,VEGF和BMP-2阳性染色细胞密度显著增高;JE组外侧区域CollagenⅢ表达显著高于内侧区域(P<0.05),外侧区域HIF-1α和VEGF表达显著高于内侧区域(P<0.01)。结论:连续跳跃运动下,兔髌骨髌腱结合部内侧与外侧区域呈现不同的损伤形态学变化特点,外侧区域Ⅲ型胶原蛋白、低氧诱导因子1α和血管内皮生长因子表达高于内侧区域。
Objective:To investigate the morphologic features and expression of related growth factor of Patella-Patellar Tendon Junction(PPTJ)injury caused by continuous jumping. Methods:Eighteen weeks old female New Zealand white rabbits were randomly divided into JE groups(JE)and control group (Con),with 5 rabbits in each group. Jumping scheme was 150 times a day,5 days a week. Patella-patellar tendon complex was harvested 4 weeks later,paraffin section were stained by HE and Safranin O to observe the histopathologic changes ,Immunohistochemistry was used to display the expression of related growth factors. Results:Compared with Con group,Cell density and Fibrocartilage zone thickness and Glycosaminoglycan area was significantly increased. Cell density in PPTJ inner zone was lower than that in PPTJ outer zone(P〈0.05),whereas Fibrocartilage zone thickness in PPTJ inner zone was larger than that of PPTJ outer zone(P〈0.05). The expression of CollagenⅢ,HIF-1α,VEGF and BMP-2 in PPTJ zone of JE group was higher than that of Con group. The expression of CollagenⅢin PPTJ outer zone of JE group was higher than that of PPTJ inner zone(P〈0.05),while HIF-1αand VEGF was higher(P〈0.01). Conclusion:Continuous jumping lead to PPTJ overuse injury,Pathological changes in PPTJ inner zone was different from that of PPTJ outer zone. The expression of CollagenⅢ,HIF-1αand VEGF in PPTJ outer zone was higher than that of PPTJ inner zone.
