健脾益肺化痰方对慢性阻塞性肺疾病大鼠胃肠动力及排痰量的影响

The Gastrointestinal Motility and Amount of phenol red expectoration Effect of Jianpiyifeihuatanfang on Chronic Obstructive Pulmonary Disease of Model Rats

目的:探讨健脾益肺化痰方对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)模型大鼠胃肠动力及排痰量的影响。方法:60只Wistar雄性大鼠随机分为正常对照组、模型组、健脾益肺方组、化痰方组、健脾益肺化痰方组和羧甲司坦组,每组10只。除正常组外,其他各组采用单纯香烟烟熏法建立COPD大鼠模型。健脾益肺方组、化痰方组、健脾益肺化痰方组和羧甲司坦组大鼠分别给予相应的药物灌胃治疗,模型组和正常组给予等体积的纯净水。于末次给药禁食12 h后,给予水合氯醛(3 m L/kg体重)皮下注射麻醉,腹主动脉取血,测定各组血清胃泌素、胃动素、生长抑素含量的变化;酚红排痰实验测定痰液的分泌情况。结果:与正常组比较,模型组大鼠血清GAS、MTL、SS含量降低(P<0.05或P<0.01),气道酚红排痰量明显升高(P<0.05);与模型组比较,各给药组大鼠血清胃肠激素含量升高(P<0.05),气道酚红排痰量均降低(P<0.01);三组中药给药组组间比较,健脾益肺方组、健脾益肺化痰方组血清胃肠激素含量进一步升高,排痰量进一步降低。结论:健脾益肺化痰方能提高大鼠胃肠运动功能,减少痰液生成,且"治本"作用优于"治标"。

Objective： To investigate the effect of Jian Pi Yi Fei Hua Tan Decoction on gastrointestinal motility and amount of phenol red expectoration drainageon chronic obstructive pulmonary disease（ COPD） of model rat. Methods： 60 male Wistar rats were randomly divided into normal control group,model group,Jianpiyifei group,Huatan group,Jianpiyifeihuatan group and Carbocisteine group,each group of 10. Inaddition to the normal control group,the model of COPD rats of other groups were established by tobacco smoke inhalation. Jianpiyifei group,Huatan group,Jianpiyifeihuatan group and Carbocisteine group were given the lavagetreatment with corresponding drugs. The model group and normal control group were given equal volume of pure water. The eating was banned for 12 hours after the last delivery. Give the chloral hydrate（ 3 ml/kg body weight） subcutaneous injection of anesthesia,blood serum were collected by abdominal aortic method,level of serum gastrin（ GAS）,motilin（ MTL） and somatostatin（ SS）. Determination of phenol red expectoration drainage experiment sputum secretion. Results： compared with normal group,model group rats serum GAS,MTL and SS content were lower significantly（ P〈0. 05 or P〈0. 01）,the phenol red expectoration drainage volume in airway of model group rats were significantly higher（ P〈0. 05）; Compared with model group,the dosage groups of rats serum GAS,MTL and SS content were increased（ P〈0. 05）,the airway row of phlegm quantity in all treating group rats were significantly lower（ P〈0. 01）. Conclusion： Jian Pi Yi Fei Hua Tan Decoction can improve airway mucus hypersecretion in COPD rats through improving gastrointestinal motor function and reducing phlegm.
