摘要
目的:研究胀果甘草多糖Gi P-B1对小鼠脾淋巴细胞增殖及诱生IL-2、IL-13、TNF-α等细胞因子的影响。方法:采用磺酰罗丹明B(SRB)法检测不同浓度Gi P-B1单独作用及其与有丝分裂原Con A、LPS共同作用对小鼠脾淋巴细胞增殖的影响;采用酶联免疫吸附实验(ELSA)法检测不同浓度Gi P-B1体外刺激对小鼠脾淋巴细胞诱生白介素-2(IL-2)、白介素-13(IL-13)、肿瘤坏死因子-α(TNF-α)等细胞因子的影响。结果:Gi P-B1在25~100μg/m L浓度范围内能显著促进小鼠脾淋巴细胞增殖,并与Con A、LPS协同促进T、B淋巴细胞增殖,还能诱导脾淋巴细胞分泌l L-2、TNF-α。结论:Gi P-B1具有免疫增强作用。
Objective: To investigate the effect of an acidic polysaccharide from Glycyrrhiza inflata( Gi P- B1) on spleen lymphocytes proliferation and induction of cytokine in mice. Methods: The effects of treatments with Gi P- B1 and with Gi P- B1 containing motigen at different concentrations on spleen lymphocyte proliferation were determined by SRB.The contents of IL- 2,IL- 13 and TNF- α were determined by ELISA. Results: Gi P- B1 at a concentration ranging from25 μg / m L to 100 μg / m L resulted in a significant increase of lymphocyte proliferation. Gi P- B1 could also enhance the effect of Con A- induced T lymphocyte proliferation and the stimulation index of LPS- induced B lymphocytes. Moreover,Gi P- B1 could increase the secretion of IL- 2 and TNF- α in spleen lymphocyte. Conclusion: Gi P- B1 has the effect on enhancing immune responses in mice.
