摘要
目的探讨胰岛素样生长因子-1(IGF-1)对人精子氧化损伤的保护作用。方法用H_2O_2处理人精子悬液建立氧化损伤模型,将精子悬液分为对照组、H_2O_2组、IGF-1+H_2O_2组。分别于培养后12 h、24 h,采用精子分析仪检测精子活力,Diff-Quik染色法进行精子形态学分析,精子染色质扩散试验检测精子DNA碎片率。结果在培养后12 h、24 h,与对照组相比,H_2O_2组精子活力及前向运动百分比均显著下降,DNA碎片率明显升高(P<0.05);IGF-1+H_2O_2组较H_2O_2组精子活力及前向运动百分比均显著升高,DNA碎片率明显下降(P<0.05);各组间正常形态精子比率无显著性差异(P>0.05)。结论 IGF-1对氧化损伤的精子具有一定的保护作用。
Objective:To investigate the protective effects of the insulin-like growth factor-1 on the oxidative injured human sperm. Methods:The sperm cells were devided into 3 groups:the control group,the H_2O_2 group and the IGF-1+H_2O_2 group.At the 12 hours or 24 hours after cultivated,the sperm motility was measured by computer assisted sperm analyzer,the sperm morphology was analyzed using the Diff-Quik staining,and the DNA fragmentaion rate was determined by sperm chromatin dispersion test. Results:Either 12 hours or 24 hours after cultivated,compared with the control group,the sperm motility and the ratio of progressive motile sperm decreased evidently and the DNA fragmentaion rate increased apparently in the H_2O_2 group(P〈0.05). However,these changes were reversed by the IGF-1 protection.There was no significant difference in the normal morphology rate among groups(P〉0.05). Conclusions:The insulin-like growth factor-1 has antioxidant effect on the human sperm.
