毛细管区带电泳法分析药桑多糖中单糖的组成 被引量:7

摘要 目的了解高效毛细管区带电泳法分析药桑多糖中单糖组成的作用。方法将药桑多糖经酸水解后,经1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化,采用高效毛细管电泳法分析各单糖组成。电泳条件:180 mmol/L硼酸缓冲液溶液,pH=10.05,柱温25℃,电压15kV,气压进样3.447 5kPa下进样10s,检测波长245nm。结果药桑多糖中主要由木糖、阿拉伯糖、葡萄糖、鼠李糖、甘露糖、半乳糖、半乳糖醛酸组成,其物质的量之比为1∶7.4∶15.8∶3.4∶10.8∶14.3∶4.5。结论高效毛细管电泳法用于多糖中单糖的组成分析具有快速、简便、精度高的特点,可用于药桑多糖中单糖组成测定和质量控制。 Objective The High Performance Capillary Zone Electrophoresis was used to analysis monosaccharide composition in polysaccharide of Morus nigra Linn.Methods The polysaccharide of Morus nigra Linn were derivatization with 1-Phenyl-3-methyl-5-pyrazolone(PMP) after hydrolyzed into monosaccharides with Trifluoroacetic Acid(TFA)and were separated by HPCE.The CE condition:Buffer:180mmol/L boric acid solution(pH 10.05),column temperature:25℃;Voltage:15kV;injection time:3.447 5kPa×10s,the detection:245nm.Results The polysaccharide was mainly composed of xylose,Arabinose,Glucose,Rhamnose,Mannose,Galactose,GalUA.The amount of the substance is 1∶7.4∶15.8∶3.4∶10.8∶14.3∶4.5.Conclusion High performance capillary electrophoresis method for the analysis of polysaccharide is rapid,simple and high accuracy.It can be used for the determination and quality control of polysaccharide in Morus nigra Linn.
