茄子嫁接愈合过程中相关酶活性的变化 被引量:6

摘要 为了明确嫁接处理后茄子嫁接苗和自根创伤苗在愈合过程中的发育差异,确定茄子嫁接愈合的关键时期,在嫁接处理后2~14d对嫁接口和创伤口间隔取样,测量茄子嫁接愈合过程生长量的变化,苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)、过氧化物酶(POD)、肉桂醇脱氢酶(CAD)活性以及木质素含量的变化。结果表明:在嫁接愈合过程中,与对照自根苗创伤口相比,嫁接苗嫁接口PAL活性和POD活性变化幅度较大;总体上嫁接口与创伤口的PPO活性变化波动不大,都呈现先上升后下降的趋势;CAD活性和木质素含量在嫁接口和创伤口的愈合过程中均表现增加趋势,但创伤口增加趋势更明显;嫁接口PPO,POD酶活性均在嫁接后第12天出现峰值,说明对于嫁接口的愈合来说第12天是重要的临界点,嫁接处理后第12天砧木与接穗的对接愈合可能基本完成;嫁接苗的生长速度慢于自根创伤苗,嫁接口愈合速度滞后于创伤口愈合速度6d。 The developmental differences of grafting union of eggplants and healing wound of own-rooted in the healing process after treatment were studied. The key period graft healing and graft healing mechanism were identified. We studied the changes of natural vegetables growth during the healing process of grafted eggplant, as well as the activities of pjenylalanine ammonia enzyme(PAL), polyphenol oxidase(PPO), peroxidase(POD), cinnamyl alcohol dehydrogenase(CAD) and content of lignin. The test materials were sampled with an 2-day interval after 2-14 day of treatment. The results indicated that the activities of PAL and POD had large variations during healing process compared with healing wound. In general, the activity of PPO of grafting union of eggplants and healing wound of own-rooted trees showed a little change, and both appeared fist rising then declining.The activity of CAD and content of lignin of the grafting union of eggplants and healing wound of own-rooted trees both performed rising tendency in the healing process, but healing wound was more dramatic. The activities of PPO, POD appeared a peak after 12 days of grafting, which illustrated that the 12 days were the critical point for the union of grafting, and rootstock and scion basically completed healing in the 12 days. The growth rate of grated seedlings was slower than that of own-rooted trees with healing wound, and the former was delayed 6 days than the later.
