Protective Effect of Trichosanthes Pericarpium on the Hyperlipemia Accompany Model Rat with Acute Myocardial Ischemia

摘要 目的:系统评估瓜蒌皮对高血脂合并急性心肌缺血大鼠的干预效果,探究瓜蒌皮"祛痰散结、开胸除痹"的现代科学内涵。方法:雄性Wistar大鼠40只,随机分为对照组,模型组,丹参片组,瓜蒌皮大、小剂量组。先采用高脂乳剂灌胃法制造高血脂模型,再结扎冠状动脉制备高血脂合并急性心肌缺血模型,对照组实行假手术。瓜蒌皮水煎液灌胃给药,丹参片作为阳性对照药。对照组及模型组灌胃生理盐水。生化试剂盒检测血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)含量,研究瓜蒌皮降血脂效果;生理记录仪记录心电图变化,评价心脏功能;心肌切片氯化三苯四氮唑(TTC)染色,测定心肌梗死面积及梗死率;试剂盒检测血浆肌酸激酶及其同工酶(CK、CK-MB)、谷草转氨酶(AST)、α-羟基丁酸脱氢酶(α-HBDH)和乳酸脱氢酶(LDH)活性,确定心肌细胞坏死程度。结果:与对照组比较,模型组大鼠血浆TC、TG、LDL显著升高,HDL降低;心电图阳性率、心肌梗死率及5项心肌酶活性均显著性升高。与模型组比较,瓜蒌皮小、大剂量组TC、TG、LDL水平均降低,HDL升高,心电图阳性率、心肌梗死率及5项心肌酶活性均显著性降低,且大剂量组效果优于小剂量组。结论:对高血脂合并急性心肌缺血大鼠,瓜蒌皮水煎液能显著降低其血脂含量,抑制缺血心肌细胞坏死,维持心脏功能。 Objective: This study aims to system assess the effectiveness of the Trichosanthes pericarpium's intervenation on hyperlipidemia accompany with coronary heart disease rat model, exploring the modern scientific connotation of Trichosanthes pericarpium about "dispel phlegm and dissipate binds, soothe the chest and diffuse impediment" . Methods : 40 Wistar male rats are randomly divided into the control group, the modle group, the danshenpian group, the Triehosanthes high-dose group and the Tnchosanthes low-dose group. We make high cholesterol accompany with acute myocardial ischemia model by iniragastrical administration with high fat emulsion and ligating the coronary artery of the Wistar rats. The control group implement the sham. the Triehosanthes decoction group and the Danshenpian group is to give intragastrical administration with Trichosanthes decoction and Danshenpian solution. The control group and the model group treat with physiological saline solution. We observe the cholesterol-lowering effect by using the biochemical diagnostic kits to detect total cholesterol ( TC ), triglyceride ( TG ), low density lipoprotein ( LDL ) and high-density lipoprotein ( HDL ) levels in plasma, detect the cardiac function by recording ECG changes with physiological recorder, evaluate the infarction rate by staining the myocardial sections with triphenyl tetrazolium chloride ( TTC ) and evaluate the degree of myocardial injury by using biochemical kits to detect levels of creatine kinase ( CK ), creatine kinase isoenzyme ( CKMB ), aspertate aminotransferase ( AST ), alpha hydroxybutyrate dehydrogenase ( c~ -HBDH ) and lactate dehydrogenase ( LDH ). Results : The level of TC/TG/LDL in the serum of the model group increased evidently and that of HDL decreased compared with the control group. The level of TC/TG/LDL of the Trichosanthes decoction group and the danshenpian group is lower compared with model group, but that of HDL is inverse, the positive rate of the ECG and myocardial infarction rate decreased obviously, also the activity of CK/CK- MB/LDH/AST/α -HBDH were remarkably lower ( P〈0.05 ). Conclusion : The Trichosanthes decoction can reduce blood lipids, decrease myocardial infarction rate of rat model with high cholesterol accompany with acute myocardial isehemia remarkedly, prevent myocardial necrosis and improve myocardial function.
