摘要
目的:研究癫痫清颗粒对β-淀粉样蛋白(Aβ_(25-35))致阿尔茨海默病模型(AD)小鼠的学习记忆改善作用。方法:60只昆明种鼠随机分为假手术组,模型组,癫痫清颗粒低(3.12 g·kg^(-1))、中(6.24 g·kg^(-1))和高(9.36 g·kg^(-1))剂量组和盐酸多奈哌齐组(1.30 mg·kg^(-1)),海马内注射(Aβ_(25-35)),制作小鼠AD模型。连续灌胃给药15 d。Y迷宫和Morris水迷宫实验检测各组小鼠认知行为改变,HE染色观察其对AD模型小鼠神经元的影响。结果:癫痫清颗粒可显著增加AD模型小鼠的自发交替反应率,缩短逃避潜伏期,减少总路程,延长游泳路程百分比,可改善海马区神经元细胞排列紊乱、水肿等现象,减少神经元死亡。结论:癫痫清颗粒对Aβ_(25-35)致阿尔茨海默病模型小鼠学习记忆能力具有改善作用。
Objective : To investigate the effects of Dianxianqing Granule on learning and memory in model mice induced by β-amyloid ( A β 25-35 ) - Methods : Sixty mice were divided into six groups randomly: sham group, model group, Dianxianqing Granule low dose group ( 3.12 g·kg^-1 ), Dianxianqing Granule medium dose group ( 6.24g·kg^-1 ) and Dianxianqing Granule high dose group ( 9.36 g·kg^-1 ) and donepezil group ( 1.30g·kg^-1 ). β -Amyloid protein ( A β 25-3s ) was infused into the hippocampal of mice. Drugs were administrated for 15 days. Y maze and Morris water maze was detected to evaluate the cognitive behavior. HE-staining was used to observe the neuron changes caused by Dianxianqing Granule. Results : Dianxianqing Granule can increase the alternation behavior significantly, decrease swimming escape latency and total distance, prolong the swimming distance percentage; it also can improve the disorder, edema and other symptoms of hippocampus neurons, reduce the death of neurons. Conclusion: Dianxianqing Granule can improve the effects of learning and memory in mice induced by β-amyloid (Aβ25-35).
