摘要
目的:观察五味子治疗维生素A缺乏型干眼症的疗效及眼角膜组织中IL-6、TNF-α的表达变化,比较泪腺细胞凋亡的数量,探讨酸味中药缓解干眼症的作用机制。方法:将180只白色来亨鸡随机分为正常组、模型组(维生素A缺乏组)、维生素A对照组、五味子治疗组(高、中、低剂量组)。分别于用药前、用药后2周、用药后4周对各组来亨鸡进行Schirmer I试验(SIT)和泪膜破裂时间(BUT)检查。对角膜做荧光素染色并用免疫组织化学检测白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α),并比较各组泪腺细胞凋亡的数量。结果:用药2周、4周后,与模型组比较,Schirmer试验滤纸湿长有明显改变,泪膜破裂时间逐渐增加,角膜表面湿润度增多,荧光素染色少量着色。免疫组织化学检测发现IL-6和TNF-α在各组来亨鸡角膜上皮细胞中表达显著降低,泪腺细胞凋亡明显缓解。结论:五味子可以提高泪液基础分泌量,维持泪膜稳定性,其机制可能是抑制泪腺细胞凋亡,抑制IL-6和TNF-α表达,减轻炎症反应。
Objective: Investigate the efficacy of Chinese medicine Schisandra to vitamin A-deficient dry eye syndrome and corneal tissue IL-6, TNF- α expression changes, compare the number of apoptotic ceils in the lacrimal gland. Methods: 180 white leghorn chicken, randomization, and start feeding. All animals were divided into normal control group, model group ( vitamin A deficient ) and .Schisandra group. The Schirmer I test ( SIT ) and tear film breakup time ( BUT ) were tested before treatment, after treatment two weeks and four weeks after treatment for each group Leghorn chickens. Fluorescein staining of the cornea and by immunohistochemistry IL-6 and TNF- α , lacrimal gland of each group number of apoptotic cells. Results : Medication two weeks, four weeks later, compared with the model group, Schirmer test paper changed significantly longer wet, tear breakup time gradually increased, increased corneal surface moisture, a small amount of fluorescein dye coloring. Immunohistochemistry detected IL-6 and TNF-α were significantly lower in the group of Leghorn corneal epithelial cells, apoptosis were relieved lacrimal gland. Conclusion : Schisandra can increase the secretion of tear, maintain the stability of tear film, its mechanism may he the inhibition of lacrimal gland cell apoptosis, inhibition of IL-6 and TNF- α expression, inflammatory reaction.
