新疆两色金鸡菊总黄酮大孔吸附树脂纯化工艺 被引量:2

摘要 目的:利用大孔吸附树脂纯化新疆两色金鸡菊中的总黄酮。方法:比较10种大孔吸附树脂对响应面优化提取所获得的新疆两色金鸡菊总黄酮的吸附和解吸性能,考察不同类型的吸附树脂对总黄酮的吸附和解吸效果。结果:AB-8型大孔吸附树脂对新疆两色金鸡菊总黄酮具有较强的吸附性能和较好的解吸能力。当总黄酮上样浓度为0.54 mg/m L,上样量为树脂体积的2.1倍(2.1 BV),吸附平衡4 h后,先用2~3 BV蒸馏水冲洗柱床,再用2.2 BV的50%乙醇洗脱,分段收集,可获得较高含量的总黄酮有效部位。结论:AB-8型大孔吸附树脂可以作为纯化新疆两色金鸡菊总黄酮的材料。 Objective: Using macroporous adsorptive resin to purificate total flavonoids from Coreopsis tinctoria Nutt. in Xinjiang. Methods : To compare the adsorption and desorption paramaters of total flavonoids with 10 different types of macroporous adsorptive resin. The total flavonoids obtained from response surface methodology ( RSM ) optimized extracts of C. tinetoria Nutt. Results : The adsorption and desorption paramaters of AB-8 macroporous adsorptive resin is better than other kinds of resins for purification total flavonoids of C. tinctoria Nutt. The high concentration of total flavonoids active fraction could obtained according optimal conditions as follows : 2.1 BV ( column bed volume, BV ) of sample solution is loaded ( the concentration of total flavonoids is 0.54 mg/mL ). After adsorption equilibrium 4 hours,2-3 BV distilled water to wash water soluble larger polar components. Then 2.2 BV 50% aqueous ethanol to elute resin bed, collection eluent. Conelution : AB-8 macroporous adsorptive resin can be used as a suitable material for purification total flavonoids from C. tinctoria Nutt. in Xinjiang.
