荸荠花粉活力测定及贮藏寿命研究 被引量:4

摘要 为了解荸荠[Heleocharis dulcis(Burm.f.)Trin.ex Hensch.]花粉生活力和最佳体外萌发条件,分别采用I2-KI、TTC、MTT染色法和离体培养基萌发法对荸荠花粉活力进行了测定,并初步研究了荸荠花粉在常温(25℃)条件和低温(4℃)贮藏条件下活力变化的动态。结果表明,MTT染色法测定结果与离体萌发法最接近,最适宜荸荠花粉活力的快速检测;离体萌发法适宜培养基配方为1%蔗糖+50μg/m L硼酸;荸荠花粉在常温条件下50 min活力降至0.29%的低水平,在低温条件下5 h后完全失去活力。 To study the pollen viability and in vitro germination conditions of Heleocharis dulcis(Burm. f.) Trin. ex Hensch.,viability of H. dulcis pollen was determined by three testing staining methods including I2-KI, TTC, MTT and in vitro culture method. Dynamic changes of in vitro pollen germination rate under the storage condition of 25 ℃ and 4 ℃ was compared.The result indicated that the pollen dyed rate of MTT staining method was similar to the in vitro germination rate, thus the MTT staining method is suitable for fast test of the pollen viability of H. dulcis. The best medium for in vitro pollen germination of H. dulcis was 1% sucrose + 50 μg / m L boric acid. The in vitro pollen germination rate of H. dulcis decreased to0.29% when stored at 25 ℃ for 50 min, while decreased to 0 when stored at 4 ℃ for 5 h.
