摘要
[目的]研究南极磷虾壳的超声辅助酶解工艺,以最大程度地回收南极磷虾加工下脚料中的蛋白质。[方法]探究了不同种类蛋白酶、固液比、p H、酶解时间、酶解温度、酶底物比以及辅助超声功率对蛋白回收率的影响,并通过正交试验优化确定超声辅助酶解工艺参数。[结果]碱性蛋白酶水解蛋白能力最强;超声辅助碱性蛋白酶解最优工艺:自然p H下,控制固液比1∶10 g/m L,辅助超声功率150 W,加入碱性蛋白酶(5 000 U/g蛋白)后,50℃下酶解2.5 h,此时蛋白质回收率达87.42%。[结论]采用优化后的超声辅助酶解工艺可极大程度地回收南极磷虾加工下脚料中的蛋白质,促进磷虾资源的充分利用。
In order to maximally recover the protein from Antarctic krill shell, the extraction technology of enzymatic hydrolysis method with Ultrasonic-assisted was studied.The influence of the different proteinases, ratio of solid-to-liquid, pH value, enzymolysis time and tempera-ture , ratio of enzyme to substrate and ultrasonic power on the recovery of protein was tested.With orthogonal experiment, the optimal process parameters were formed, which was as follows:the pH value was in natural condition; the ratio of solid to liquid, 1∶10 g /mL; the ultrasonic power, 150 W; the ratio of enzyme to substrate, 5 000 U /g protein; the temperature, 50 ℃; and the treating time, 2.5 h at 50 ℃.The ex-traction efficiency of protein in the optimized conditions was 87.42%.Alkaline protease had the strongest capability in hydrolyzing protein.As a result, the protein in Antarctic krill shell could be recovered with the technique to the most extent and the kinds of resource could be fully u-tilized.
