Simultaneous Detection of 13 Kinds of Polyphenols in Fruit Wine by Reversed-Phase High Performance Liquid Chromatography

摘要 建立了一种适用于多种果酒中13种多酚类化合物(5种黄烷-3-醇、4种黄酮醇、2种酚酸、2种芪类)含量测定的HPLC分析方法。色谱条件为:Li Chrospher 100 RP-18e柱,流动相A为水(0.1%磷酸),流动相B为甲醇(0.1%磷酸),梯度洗脱,流速为0.5 m L/min,检测波长为272 nm。在该色谱条件下各个物质在48 min内得到了良好的分离,各物质的含量与峰面积呈良好的线性关系,相关系数(R2)均大于0.998,且回收率在73%~123%之间,重复性好,具有定量准确、快速等特点。运用该方法测定了6种葡萄酒、7种蓝莓酒、5种桑葚酒中13种酚类化合物的含量。结果表明,葡萄酒中含有全部13种多酚化合物,蓝莓酒中有10种,桑葚酒中有9种。葡萄酒中黄烷-3-醇的含量较高,蓝莓酒和桑葚酒中黄酮醇的含量较高,葡萄酒和桑葚酒均含有白藜芦醇,蓝莓酒不含。该方法准确、可靠,可适用于多种果酒中13种酚类化合物的测定。 A new method of RP-HPLC has been developed for simultaneous detection of 13 polyphenols (5 flavan-3-ols, 4 flavonols, 2 pheno-lic acids and 2 stilbene) in fruit wine. The separation was performed on a LiChrospher 100 RP-18e column at 30℃. The mobile phase was con-sisted of 0.1%phosphoric acid (A) and 0.1%phosphoric acid methanol (B), with gradient elution at a flow rate of 0.5 mL/min. The injection volume was 10μL and a Photo-Diode Array (PDA) was used to monitor signals at 272 nm. Under these chromatographic conditions, 13 poly-phenols were successfully separated in 48 minutes and their contents showed good linear relationships with the peak areas, R2〉0.998. The method had the advantages of high recovery rate(73%~123%), good repeatability, good accuracy, and rapid detection. The contents of 13 polyphenols in six grape wine products, seven blueberry wine products, and five mulberry wine products were determined by this method. The results suggested that, 13, 10, and 9 polyphenols could be detected in grape wine, blueberry wine, and mulberry wine, respectively;the content of flavan-3-ols in grape wine was higher, on the other hand, the content of flavonols in blueberry wine and mulberry wine was higher;resvera-trol was detected in grape wine and in mulberry wine, while there was no resveratrol in blueberry wine. This mehtod was suitable for the detec-tion of mutiple phenolic compounds in various wine products.
