摘要 【目的】为了优化板栗花粉的液体培养体系,研究蔗糖、硼酸和氯化钙的浓度等因素对花粉萌发率的影响。【方法】以‘燕红’为材料,在蔗糖、硼酸、氯化钙单因子试验的基础上,采用正交试验设计筛选板栗花粉液体培养的最适条件。【结果】研究结果表明:最终筛选的板栗花粉最适液体培养条件为:15%蔗糖、0.005%硼酸和0.04%氯化钙,最适pH值为6.5,培养温度为34℃,转速为175r/min。在此条件下,保存一年的花粉萌发率仍可达到28.86%。【结论】说明板栗花粉的萌发率受培养基组分、pH、转速和培养温度等条件影响。 [Objective]To optimize the liquid culture system of Chinese chestnut (Castanea mollissima Blume. ) ('Yanhong') pollen, we investigated the effects of the concentration of sucrose, H3 BO3. CaCl2 and other factors on pollen germination rate. [Methods]The single factor such as sucrose, H3BO3 and CaCl2 was tested. Based on the above result, orthogonal experiment was carried out to optimize the liquid culture condition for Chinese chestnut pollen. [Results]The results showed that the medium contained 15% sucrose, 0. 005% H3BO3, 0.04% CaCl2, pH 6.5, 34℃ and 175 r/min were optimal for the pollen germination of Chinese chestnut. Under the above cultural condition, the pollen germination rate reached 28.86%, even the pollens had been preserved for one year. [Conclusion]The results indicated that pollen germination rates were mainly affected by several factors such as medium components, pH value, rotation speed and culture temperature.
