摘要
目的:研究芹菜素诱导人膀胱癌5637细胞凋亡的作用机制。方法:采用25~100μmol/L芹菜素处理膀胱癌5637细胞,流式细胞仪检测细胞凋亡情况;采用Western blotting检测细胞凋亡相关信号分子Bax、Bcl-2和PARP蛋白的表达水平。结果:芹菜素处理使细胞Bcl-2蛋白表达降低且呈浓度依赖性,并使Bax的表达增加和导致PARP蛋白发生切割且呈浓度依赖性。结论:芹菜素能够抑制人膀胱癌5637细胞的增殖,诱导细胞凋亡,其机制可能与下调Bcl-2并上调Bax表达,导致Bcl-2/Bax比值下降,PARP活化有关。
Objective To study the mechanism of apigenin on the apoptosis of human bladder cancer cell line 5637.Methods 5637 cells were cultured with different concentrations of apigenin,and MTT assay was used to evaluate the cell inhibition rates.The expressions of apoptosis-related proteins Bax,Bcl-2 and PARP were analyzed by Western blotting.Results Apigenin causes concen-tration -dependent inhibition of the proliferation of bladder cancer 5637 cells.Western blotting demonstrated that apigenin increased the protein levels of Bax and PARP cleavage and reduced the protein expression of Bcl-2.Conclusion Apigenin can inhibit the prolif-eration and induce apoptosis of 5637 cells possibly by up-regulating the Bax expression,enhancing PARP cleavage,downregulating the Bcl-2 expression and resulting in the Bcl-2/Bax ratio decreased.
