摘要
目的观察糖尿病模型大鼠脊髓前角中凋亡相关基因Bax与Bcl-2表达变化及电针对两者表达的影响,探讨电针改善糖尿病神经系统损伤的可能机制。方法 32只雄性Wistar大鼠随机分为正常组、溶剂组、模型组和电针组,每组8只。模型组和电针组腹腔注射链脲佐菌素制备糖尿病大鼠模型。电针组于糖尿病造模成功1周后取"足三里""胰俞"穴给予电针刺激,每次治疗30min,每天1次,共4周。观察造模后及治疗后各组大鼠空腹血糖水平;用免疫组织化学染色方法检测脊髓前角的Bax与Bcl-2的蛋白表达。结果与正常组相比,模型组血糖水平显著升高,脊髓前角Bax蛋白表达明显增加(P<0.05),而Bcl-2蛋白表达则明显减少(P<0.05);与模型组相比,电针组大鼠血糖水平明显降低,脊髓前角Bax蛋白表达明显降低(P<0.05),而Bcl-2蛋白表达则明显升高(P<0.05);电针组与溶剂组和正常组大鼠的Bax和Bcl-2蛋白表达水平相接近(P>0.05)。结论糖尿病模型大鼠脊髓前角神经元凋亡增加;电针可明显拮抗糖尿病模型大鼠脊髓前角神经元的凋亡,从而改善糖尿病大鼠脊髓前角损伤。
Objective To observe the changes of apoptosis related gene Bax and Bcl-2 expressions in an-terior horn of spinal cord in diabetic rat models and the effects of electroacupuncture (EA)on both Bax and Bcl-2,so as to explore the possible mechanism of electroacupuncture in improving the damage of diabetic nervous system. Methods Thirty-two male Wistar rats were randomly divided into a normal group,a vehicle group,a model group and an EA group (each group n=8).The rat models of diabetes (the model group and the EA group)were established by intraperitoneal inj ection a dosage of streptozotocin.One week after the suc-cessful establishment of diabetic rats,the EA group rats were given electroacupuncturing “Zusanli”and“Yishu”points for 30 minutes,once daily for 4 weeks.After the diabetic rats being successfully established and at the end of EA treatment,the fasting blood glucose levels of all rats were observed;and an immunohisto-chemical method was used to detect Bax and bcl-2 protein expressions of anterior horn of spinal cord. Re-sults In comparison with the normal group,the blood glucose levels and the protein expression of Bax in ante-rior horn of the spinal cord in the model group significantly increased (P〈0.05),but the expression of Bcl-2 protein significantly decreased (P〈0.05).In comparison with the model group,the blood glucose levels and the Bax protein expression of anterior horn of spinal cord in EA group remarkably decreased (P〈0.05),but the expression of Bcl-2 protein significantly increased (P〈0.05),and expressions of Bcl-2 and Bax proteins of EA group were close to the vehicle group and the normal group (P〉0.05). Conclusion The neuron ap-optosis of anterior horn of spinal cord in diabetic rat models were increased;EA could significantly antagonize the neuron apoptosis of anterior horn of spinal cord in diabetic rat models,so as to improve the anterior horn inj ury of diabetic rats.
