不同组培方法对香蕉组培苗假植阶段生理特征的影响 被引量:3

Effect of Different Tissue Culture Methods on Physiological Characteristics of Banana Plantlets at Transplanting Stage

摘要 以培养获得的第6代香蕉不定芽为试验材料,以两种组培方式(间歇浸没式生物反应器TIBs、传统半固体培养基)培养香蕉组培苗,进行假植移栽后20、40、60 d,分别测定不同组培苗叶片的光合特性及抗氧化酶活性等指标,比较两种组培方式对香蕉组培苗假植阶段的光合生理的影响,分析TIBs系统培养的香蕉组培苗对自然环境的适应能力。结果表明,随着移栽天数的增加,两种香蕉组培苗叶片光合速率、气孔导度等逐渐增加,而胞间CO2浓度逐渐下降,其中以TIBs培养的香蕉组培苗表现较高;两种香蕉组培苗叶片叶绿素含量及SOD、POD活性均随移栽天数的延长而上升,均以TIBs方式培养的组培苗较高;叶片MDA含量随移栽时间的延长而降低,以传统培养苗表现较高。说明在移栽后60 d内,TIBs培养的香蕉组培苗光合作用、养分吸收能力及抗逆能力均优于传统半固体培养的组培苗,TIBs可作为香蕉组培苗快繁的有效方法。 The adventitious buds derived from the 6thgeneration banana plantlets were used as tested materials to induce new plantlets by using two tissue culture methods viz.,Temporary Immersion Bioreactor system( TIBs) and traditional semi-solid medium culture. The plantlets were transplanted to heel in cultivating bag with 70 % sandy soil and 30 % leaf mould. The photosynthetic characteristics and antioxidase activities in leaves of different banana plantlets for two culture methods were investigated after 20,40 and 60 days,in order to compare the influences of two culture methods on photosynthetic physiology of plantlets and analyze the environmental adaptability of plantlets derived from TIBs culture method, respectively. The results showed that,the net photosynthesis rate,stomatal conductance,transpiration rate in plantlet leaves of the two culture methods were gradually increased as the time goes on,but the intercellular CO_2 concentration in leaves was declined; of which,the plantlets derived from TIBs culture method presented more higher value. That was means,the photosynthetic performance of banana plantlets for TIBs method was much better than that of semi-solid culture. The SOD and POD activities and chlorophyll content in leaves of the two culture methods were also increased with the elongation of transplanting time,and the higher one were found in leaves of plantlets by using TIBs culture method; while the MDA content was declined,and the higher value was found in leaves of plantlets by using semi-solid culture method. The above-mentioned results indicated that,banana plantlets which derived from TIBs performed higher photosynthesis ability,nutrient absorbing ability and stress resistance during transplantation stage,and TIBs is an efficient method for banana tissue culture plantlets rapid propagation.
