摘要
[目的]探索配合物对DNA的断裂作用,寻找新型高效的核酸断裂试剂。[方法]设计合成了一例未见文献报道的基于喹啉骨架的二羧酸单核锌(Ⅱ)配合物[ZnL(H_2O)_3]·H_2O。运用元素分析、红外吸收光谱方法表征了该配合物,并利用X-射线单晶衍射仪解析其单晶结构,利用琼脂糖凝胶电泳手段研究了其化学核酸酶活性。[结果]解析结构表明配合物中Zn(Ⅱ)中心为七配位的畸变五角双锥结构;配合物表现出一定程度的切割DNA活性。[结论]本研究为设计合成新型高效、结构简单的化学核酸酶提供有价值的研究基础。
[Objective]In order to further explore the DNA cleavage of new and efficient nucleic acid fracture reagent.[Methods]a new mononuclear Zinc(Ⅱ)complex [ZnL(H_2O)_3]·H_2O of quinoline derivatived ligand(L=sodium 8-(carboxylatomethoxy)quinoline-2-carboxylate has been synthesized).The conpler was characterized using element analyses and infrared spectroscopy.The structure of the complex has been determined by X-ray diffraction crystallographic method.[Results]The Zn(Ⅱ)center was heptacoordinated with O_6N donor sets and the geometry around metal center can be best described as distorted pentagonal bipyramidal.The nuclease activity of the complex has been investigated by gel electrophoresis,and the result showed that complex exhibited a certain chemical nuclease activity.[Conclusion]This study would be beneficial to design efficient and simple structural chemical nuclease.
