摘要
目的:研究超声法提取山茱萸多糖的最佳工艺,并考察初步纯化后山茱萸多糖对人肝癌HepG2细胞的抗肿瘤活性。方法:采用正交法优化山茱萸多糖的超声提取工艺;用MTT法考察经脱蛋白、脱色纯化后山茱萸多糖对人肝癌HepG2细胞的抑制作用。结果:山茱萸多糖的最佳超声提取工艺为:水料比20∶1、超声功率230W、超声时间40min、提取温度70℃;山茱萸多糖作用后体内肝癌HepG2细胞的生长受到明显抑制,细胞增殖抑制率呈明显的量效、时效关系。结论:该山茱萸多糖超声提取工艺简单易行、重现性好,且山茱萸多糖可抑制人肝癌HepG2细胞生长。
Objective. To optimize the ultrasonic extraction process of polysaccharides from Comus officinalis. To study the antitumor activity of polysaccharides from Comus officinalis on human hepatocellular carcinoma HepG2 cells. Methods: The orthogonal design was applied to optimize the ultrasonic extraction process of polysaccharides from Comus officinalis. The inhibitory effects of the purified polysaccharides from Comus officinalis by deproteinated process on human hepatocellular carcinoma HepG2 cells were researched by MTT method. Results: The ultrasonic extraction process of polysaccharides from Comus officinalis resulted from the experiment is simple and reproducible. Conclusion : The polysaccharides from Comus officinalis can inhibit the growth of human hepatocellular carcinoma HepG2 cells.
