摘要
目的研究再生蛋白-1被干涉后对胃癌细胞株MKN45侵袭转移能力的影响。方法通过实时定量PCR、Western blot检测再生蛋白-1在正常胃黏膜细胞与不同胃癌细胞株中的表达水平;通过Transwell细胞转移实验、Transwell细胞侵袭实验检测再生蛋白-1对胃癌细胞株MKN45侵袭转移能力的影响。结果再生蛋白-1在胃癌细胞株中的表达水平明显高于正常胃黏膜细胞;当再生蛋白-1被干涉后,胃癌细胞株MKN45的侵袭转移能力明显下降。结论再生蛋白-1能够促进人类低分化胃腺癌细胞株MKN45的侵袭转移能力。
Objective To study the effect of neogenin-1 knockdown on the invasion and metastasis of gastric cancer cell line MKN45. Methods The expression level of neogenin-1 in the normal gastric mucosa cells and different gastric cancer cell lines were detected by Western blot and quantitative real-time PCR( qRT-PCR). The effect of neogenin-1 on the migration and invasion ability of gastric cancer cell line MKN45 was examined by using the transwell chamber assay and wound healing assay. Results The expression level of neogenin-1 was much higher in gastric cancer cell lines than that in normal gastric mucosa cells. The migration and invasion ability of gastric cancer cells was significantly decreased after neogenin-1 knockdown. Conclusion Neogenin-1 can increase the migration and invasion ability of poorly differentiated gastric cancer cell line MKN45.
