深绿木霉T2发酵液蛋白提取物TraT2A诱导兰州百合抗灰霉病研究 被引量:9

Resistance of Lanzhou Lily to Botrytis cinerea Induced by Protein Extract Tra T2A From Trichoderma atroviride T2 Fermentation Liquid

摘要 为了探讨深绿木霉T2发酵液蛋白提取物Tra T2A诱导兰州百合对灰霉病菌产生抗性,采用不同浓度的Tra T2A测定了对百合灰霉病菌的抑菌率和诱导抗病作用,其后用Tra T2A的100倍液喷雾处理兰州百合,48 h后接种灰霉菌,研究其诱导抗病机理。结果表明:10倍、100倍Tra T2A对百合灰霉菌有一定的抑制作用和诱导抗病作用,100倍液的抑菌率最小,仅为1.16%,但其诱导抗病效果最大,为84.77%。诱导处理Tra T2A+接种灰霉菌能显著提高兰州百合叶片中GLU、PAL、POD、PPO活性及蜡质含量并促进胼胝质的积累,其中该处理后的POD、PPO活性和蜡质含量在测定时间内均高于处理Tra T2A、接种灰霉菌和CK,GLU活性除第3天外,其余时间也高于其他3个处理。GLU、PAL活性在第7天达到最大值,分别为284.28、2.93 U,比其他3个处理分别高出0.09、0.31、0.46倍和0.31、0.19、0.47倍;POD、PPO活性和蜡质含量在第1天达到最大值,分别为1.59 U、0.63 U、14.48 mg/g,比其他3个处理分别高出0.04、0.29、0.43倍,0.12、0.32、0.67倍和0.09、0.06、0.99倍;胼胝质积累也均高于其他各处理。说明Tra T2A诱导处理能够激活百合防御体系,使百合产生抗病性。 In order to induce the resistance of Lanzhou lily against Botrytis cinerea, different concentrations ofprotein extract Tra T2 A from Trichoderma atroviride T2 fermentation liquid were added to the culture medium.The antibacterial rate and the induced resistant effect to Botrytis cinerea were investigated. To study theinduced disease resistant mechanism, the 100 times diluent of Tra T2 A was sprayed to healthy Lanzhou lily,and Botrytis cinerea was inoculated after 48 hours(Tra T2A+IB), and the other three treatments were Tra T2 A, IB(inoculating Botrytis cinerea) and CK(the control group). The results indicated that the 10 times and 100 timesdiluent of Tra T2 A had antibacterial effect on Botrytis cinerea, and the antibacterial rate of 100 times diluent ofTra T2 A was the minimum, which was only 1.16%, but the induced resistant effect was the maximum, which reached 84.77%. The treatment of Tra T2A+Botrytis cinerea significantly improved the activities of GLU, PAL,POD, PPO, wax content and the accumulation of callose. And the activities of POD, PPO and the wax contentwere higher than that of other treatments. The activity of GLU was higher than that of other treatments except inthe third day. The activities of GLU and PAL reached the maximum in the seventh day, which were 284.28 and2.93 U, respectively, and was 0.09, 0.31, 0.46 times and 0.31, 0.19, 0.47 times higher than that of Tra T2 A, IBand CK treatment, respectively. Meanwhile, the activities of POD and PPO and the max content all reached themaximum in the first day, which were 1.59 U, 0.63 U and 14.48 mg/g, respectively. The activity of POD in thetreatment of Tra T2A+IB was 0.04, 0.29, 0.43 times higher than that of the other three treatments, the activity ofPPO was 0.12, 0.32, 0.67 times and the wax content was 0.09, 0.06, 0.99 times higher than that of the otherthree treatments. And the accumulation of callose was also higher than that of the other three treatments. Theresults indicated that the treatment of Tra T2A+IB could activate the defense system of lily and induce diseaseresistance.
