摘要
本研究旨在筛选适宜花生上胚轴愈伤组织诱导的最佳培养基,建立并优化花生上胚轴愈伤组织诱导体系。实验选用不同类型的花生品种,以上胚轴为外植体,在添加有3~6 mg/L Pic和5~10 mg/L2,4-D的MS-B5诱导培养基上对上胚轴愈伤组织诱导。比较不同预培养时间的愈伤组织诱导差异,不同基因型诱导差异以及不同激素配比诱导差异。结果表明,预培养0天的珍珠豆型花生‘白沙1016’诱导率最高,确定MS+B5+Pic 3 mg/L为最适宜诱导培养基,MS+Pic 3 mg/L+Gln 1 g/L为继代培养基。该体系适宜本试验中多数花生品种的愈伤诱导,并可进行多次继代培养得到重复性体胚发生培养物。
The objectives were to screen the optimal medium for inducing the epicotyls callus of peanut,establish and optimize the callus-induction system of peanut epicotyls. Different types of peanut varieties wereused in this experiment. The epicotyls were used as explants. The epicotyls were cultured on MS-B5 mediumsupplemented with 3-6 mg/L Pic and 5-10 mg/L 2, 4-D to induce callus. The induction differences of calluswere compared with different preculture time, different genotypes, and different plant hormones concentrations,respectively. The results showed that Spanish type‘Baisha 1016'with preculture for 0 day had the biggestinductivity, the optimal medium was MS+B5+Pic 3 mg/L, the subculture medium was MS+Pic 3 mg/L+Gln1 g/L. This system is suitable for the induction of most peanut varieties and repetitive somatic embryogenesis can be obtained by repeated subculture.
