摘要
将20只鸡随机分成对照组和试验组,对照组单剂量灌胃给予左氧氟沙星(levofloxacin,LVLX)10mg/kg;试验组左氧氟沙星10mg/kg与甘草提取物0.3g/kg合用。采用HPLC法测定血药质量浓度,并以DAS2.0软件计算并比较主要药动学参数。结果显示:对照组和试验组左氧氟沙星的主要药动学参数C_(max)分别为(1.415±0.116)、(1.321±0.165)mg/L,t_(max)分别为(1.983±0.172)、(2.365±0.325)h,AUC_(0~36h)分别为(8.659±0.852)、(9.415±1.176)mg·L^(-1)·h^(-1),t_(1/2)分别为(3.714±0.674)、(8.341±0.853)h,MRT_(0~36h)分别为(4.116±0.238)、(6.648±0.676)h,Vz/F分别为(3.154±0.296)、(3.315±0.436)L/kg。与对照组相比,试验组药动学参数C_(max)降低,AUC_(0~36h)、Vz/F增大,但差异均不显著(P>0.05);t_(max)、t_(1/2)、MRT_(0~36h)均极显著增大(P<0.01)。结果表明:合用甘草提取物和左氧氟沙星,甘草提取物能减慢左氧氟沙星在体内的吸收和消除速度,延长药物在体内的作用时间。
Twenty experimental chickens were randomly divided into control and test groups.The chickens in control group were orally given a single dose of levofloxacin at 10mg/kg,the chickens in test group were orally given a single dose of levofloxacin at 10mg/kg and licorice extract at 0.3g/kg simultaneously.The concentrations of levofloxacin in plasma were determined by high performance liquid chromatography.The pharmacokinetic parameters between two groups were compared by DAS2.0program.The main pharmacokinetic parameters in chickens were as follows respectively,control group:Cmax=(1.415±0.116)mg/L,tmax=(1.983±0.172)h,AUC0-36h=(8.659±0.852)mg/L·h,t1/2=(3.714±0.674)h,MRT036h=(4.116±0.238)h,Vz/F=(3.154±0.296)L/kg;test group:Cmax=(1.321±0.165)mg/L,tmax=(2.365±0.325)h,AUC0-36h=(9.415±1.176)mg·L^(-1)·h^(-1),t_(1/2)=(8.341±0.853)h,MRT_(0-36h)=(6.648±0.676)h,Vz/F=(3.315±0.436)L/kg.Compared with the main pharmacokinetic parameters of LVLX,Cmaxin test group was lower than that in control group(P〈0.05),AUC_(0-36h),Vz/F in test group were higher than that in control group(P〈0.05),but t_(1/2),tmax,MRT_(0-36h) was significantly higher than that in control group(P〈0.01).The results showed that licorice extract could influence the pharmacokinetic parameters of levofloxacin in chickens significantly,including decreasing the speed of absorption,retarding the speed of elimination and lengthening the time of drug action.
