摘要
目的:采用正交试验设计法优化龙胆中龙胆总苷的提取工艺。方法:通过预实验和单因素试验,最终选用回流提取法,并以乙醇浓度、乙醇的体积、提取时间为自变量,龙胆总苷的百分含量为因变量,采用L9(34)正交试验设计最终确定龙胆的最佳提取工艺。结果:龙胆的最佳提取工艺为:60%的乙醇提取3次,乙醇用量为12倍,提取时间为第1次2小时,第2次1小时,第3次1小时。龙胆总苷的百分含量能达到8.73%。结论:正交试验设计法优化龙胆中龙胆总苷的提取工艺,环保,能耗低,方法简便,提取效率高,预测性良好。
Objective: To optimize the extraction technology of total gentiin in Longdan using orthogonal design method. Method: Pre-test and one-factor test were carried to optimize the technology and reflux extraction was selected. Ethanol concentration, ethanol volume and extraction time were used as independent variables, the percentage of total gentiin was used as the dependent variable. L9(34) orthogonal experimental design was applied to ultimately determine the best extraction process. Result: The optimal extraction process of gentian was as follows: 60% ethanol, extract three times(2h, lh, and lh each time), 12 times of ethanol. Total gentiin percentage was 8.73%. Conclusion: The extraction process of total gentian optimized by orthogonal design is environmental with low energy consumption. The method is simple, and the extraction efficiency is high with good predictability.
