电针对控制性超排卵小鼠子宫内膜IGF-1的影响

Effect of acupuncture on IGF- 1 in the endometrium of COH mice

目的观察子宫内膜IGF-1在电针改善控制性超排卵(COH)小鼠妊娠率中的作用。方法将昆明纯种性成熟雌性小鼠随机分成自然周期组、COH组和电针组,每组又分为供体鼠和受体鼠,每组20只。采用控制性超促排长方案制备动物模型,并行胚胎移植,于HCG注射日行电针治疗,观察临床妊娠率和胚胎着床位点数,并采用Western blot和RTPCR检测子宫内膜IGF-1蛋白及其mRNA表达。结果 COH组妊娠率、平均着床位点数和子宫内膜IGF-1蛋白及其mRNA表达显著低于自然周期组(P<0.01);电针治疗后,表达增加,且妊娠率、平均着床位点数显著提高(P<0.05)。结论在IVF-ET治疗不孕症过程中,配合针刺可促进子宫内膜分泌IGF-1,从而提高临床妊娠率。

Objective To observe the effect of IGF- 1 in the endometrium in improving the pregnancy rate of COH mice. Methods Randomly divided the mature female mice into the natural cycle group（ NC）,controlled ovarian hyperstimulation（ COH）group,electro- acupuncture group（ EA）. Each group was also divided into the donor and recipient rats,20 in each group. The COH mice model was prepared by controlled ovarian hyperstimulation,and made embryo transplantation,EA were treated with electroacupuncture on HCG injection day,observed the pregnancy rate and the number of implantation sites,detection of IGF- 1and mRNA expression in endometrium with RT- PCR and Western blot. Results Compared with the natural cycle group,the pregnancy rate,the average number of implantation sites,IGF- 1 and mRNA were significantly lower in the COH group（ P〈 0.01）; After electroacupuncture treatment,the pregnancy rate,average number of implantation sites IGF- 1 and mRNA were significantly higher than that in the COH group（ P 〈0. 01）. Conclusion In the treatment of infertility by acupuncture combined with IVF- ET,can increase the secretion of IGF- 1 in the endometrium,so as to improve the pregnancy rate.
