固定化高效石油降解菌处理石油废水优化实验 被引量:2

摘要 通过采集石油污染土壤,进行富集、培养、分离纯化,选取其中1株石油降解菌(CQ-5),采用长庆油田所采原油为降解底物,以核桃壳为固定化载体,采用吸附法固定微生物,分别考察pH值、温度、原油初始浓度、固定化时间对原油降解率的影响;通过响应面分析法优化降解条件,在此基础上进行降解实验.结果表明,单因素对原油降解率影响大小依次是温度、pH值、原油初始浓度、固定化时间,根据响应面模型分析确定最佳降解条件为pH值7.2、温度32℃、原油浓度为3 898mg/L、固定化时间为29.8h,此时7天原油降解率达到66.8%. CQ-5,A strain of oil degrading bacterium(CQ-5)was obtained from the oil contaminated soil through its enrichment,culture,isolation and purification.Taking the Changqing crude oil as substrate and walnut shells as immobilization carrier,the embedding method was adopted to immobilize microorganism and the influence of pH value,temperature,initial concentration of crude oil and immobilized time on crude oil degradation rate was explored.The degradation condition was optimized with response surface analysis method and based on it,the degradation experiment was conducted.The result showed that the influence degree of single factor on degradation of crude oil in a descending order will be temperature,pH value,initial concentration of crude oil and immobilized time.According to the analysis with response surface model,the optimal degradation conditions will be as follows:pH value of 7.2,temperature of 32 ℃,initial concentration of crude oil of 3 898mg/L and the immobilized time of 29.8h.The degradation rate of crude oil will be up to 66.8%in such case.
