摘要
为探讨抗氧化剂褪黑素(MLT)对牛卵母细胞体外成熟及胞浆单精子注射(ICSI)显微操作后早期胚胎体外发育的影响。在牛卵母细胞体外成熟培养液中,分别添加0 M、10^(-10) M、10^(-9) M、10^(-8) M MLT,通过统计核成熟率,评估不同浓度的MLT对牛卵母细胞体外成熟的影响。于24h后收集核成熟的牛卵母细胞进行显微操作,同时在显微操作液中分别添加0 M、10^(-10) M、10^(-9) M、10^(-8) M MLT,经体外培养后评估MLT对ICSI处理后胚胎体外发育的影响。结果表明:与对照组相比,在成熟液中添加10^(-9) M MLT(73.8%vs 81.3%)(P<0.05)可显著提高牛卵母细胞成熟率;同时在显微操作液中添加10^(-9) M MLT,与对照组相比,牛卵母细胞ICSI后胚胎体外发育的卵裂率(81.3%vs 85.5%)差异不显著(P>0.05),但囊胚率(14.3%vs 22.6%)显著提高(P<0.05)。因此,添加10^(-9) M MLT有助于优化ICSI技术体系,提高显微授精后胚胎体外生产效率。
The effect of antioxidative MLT on bovine oocyte maturation and embryo development in vitro after ICSI micromanipulation was investigated.After 24 h,mature oocytes were collected in vitro and maturation medium were supplied with four concentrations of melatonin(10^-10 M,10^-9 M,10^-8 M,0M).And then the effect of different concentrations MLT on bovine oocyte maturation in vitro by PB1 emission was evaluated.The matured bovine oocytes were collected to be used in micromanipulation,at the same time,MLT(0M,10^-10 M,10^-9 M,10^-8 M)was added in the micromanipulation liquid to evaluate the effect of different concentrations of MLT on ICSI embryo development in vitro.The results showed that supplementation with MLT of 10^-9 M in the culture medium resulted in a higher(P〈0.05)maturation rate(73.8% vs 81.3%).Compared with control group,the cleavage rate of ICSI embryos in the micromanipulation liquid which was added 10^-9 M MLT has no significant difference to other groups(73.8%vs 81.3%,P〈0.05);however,comparing with the control,the blastocyst rate of that was significantly higher(14.3% vs 22.6%,P〈0.05).Above all,the results help to optimize ICSI procedures and improve ICSI embryo development in vitro.
