齐墩果酸通过激活Nrf2/HO-1信号通路减轻小鼠肝脏缺血-再灌注损伤 被引量:1

Oleanolic acid alleviates hepatic ischemia-reperfusion injury by activating Nrf2/HO-1 signaling pathway in mice

摘要 目的探讨齐墩果酸(OA)对小鼠肝脏缺血-再灌注(I-R)损伤的作用及可能机制。方法取24只小鼠建立部分肝脏I-R模型后均分为四个实验组:I-R组(模型对照)、OA组(OA预处理)、二价锌原卟啉Ⅸ(ZnPPⅨ)组(ZnPPⅨ预处理)、OA+ZnPPⅨ组(OA+ZnPPⅨ预处理)。另外取24只小鼠作为I-R模型的假手术对照(S)组。实验组小鼠再灌注6h后处死。检测各组血清ALT、肝组织丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性,Western blot检测核转录相关因子2(Nrf2)和Ⅰ型血红素氧化酶(HO-1)的表达。结果与S组比较,I-R组血清ALT和MDA水平升高(P<0.05),SOD活性降低(P<0.05)。与I-R组比较,OA组ALT和MDA水平降低(P<0.05),SOD活性升高(P<0.05),Nrf2和HO-1表达升高(P<0.05)。与OA组比较,OA+ZnPPⅨ组HO-1蛋白表达降低(P<0.05),血清ALT和MDA水平升高(P<0.01或P<0.05),SOD活性降低(P<0.01)。结论 OA预处理可通过激活Nrf2/HO-1信号通路减轻小鼠肝脏I-R损伤。 Objective To investigate the effect and underlying mechanism of oleanolic acid (OA) on liver ischemia-reperfusion(I-R) injury of mice. Methods After establishmen of mice model with partial warm hepatic I-R injury, 24 mice were equally assigned into 4 test groups of I-R(model control), OA (pre-treated with OA), bivalent zinc protoporphyrin Ⅸ (ZnPP Ⅸ ) (pre-treated with ZnPPIX) and OA+ZnPPⅨ(pre-treated with OA plus ZnPPⅨ). Another 24 mice were taken as the sham-operated controls (group S). The mice of four test groups were sacrificed after reperfusion for 6 hours. The levels of ALT, MDA and SOD were measured and the expressions of nuclear factor- related factor 2(Nrf2) and hemoglobin oxidase type I (HO-1) were detected by Western blot. Results Compared with group S, the mice in group I-R had significantly higher levels of serum ALT and MDA,but lower activity of SOD (P〈0. 05). Compared with group I-R, the mice in group OA had significantly lower levels of serum ALT and MDA, but higher activity of SOD(P〈0. 05). Compared with group I-R, the mice in group OA had up-regulated expressions of HO-1 and Nrf2(P〈0. 05). Compared with group OA, the mice in group OA+ZnPPⅨ had down-regulated expression of HO-1, higher levels of serum ALT and MDA,and significantly lower activity of SOD(P〈0.01 or P〈0. 05). Conclusion OA alleviates hepatic I-R injury by activating the Nrf2/HO-1 signaling pathway in mice.
