Wee1抑制剂联合阿霉素对急性白血病细胞的生长抑制作用

摘要 目的:研究应用Wee1抑制剂PD0166285联合阿霉素对急性白血病细胞的生长抑制。方法:以人急性白血病细胞株HL60、NB4、EOL-1为研究对象,采用四甲基偶氮唑盐比色法(MTT法)检测PD0166285和阿霉素对细胞的生长抑制作用。结果:单独用药实验结果示,PD0166285或者阿霉素对HL60、NB4、EOL-1均有不同程度的生长抑制作用,并呈明显的浓度一效应关系。联合用药实验结果显示,PD0166285和阿霉素联用后对HL60、EOL-1细胞株的抑制率显著增加,明显高于单一用药,且有显著差异(P<0.05),对NB4细胞株的抑制率也增加,但无显著差异(P>0.05)。结论:相对于单独应用PD0166285或者阿霉素,PD0166285联合阿霉素增强了对急性白血病细胞株HL60、EOL-1的生长抑制作用。 Objective: To investigate the effects of Wee1 inhibitor PD0166285 and Doxorubicin induce growth arrest of AML cell lines by MTT assay. Methods: Taking human AML cell line HL60,NB4,EOL- 1 as the research objects,we investigated the inhibition of PD0166285 and Doxorubicin on cell growth by MTT Assay. Results: The results of solo- medication group showed that PD0166285 and Doxorubicin inhibited the growth of HL60、NB4、EOL- 1 cells in a dose- dependent manner. The results of combined- medication group showed that PD0166285 and Doxorubicin which used in combination inhibited the growth of HL60、EOL- 1 cells. The inhibition was more obvious in the combination group than in the solo- medication group(P〈0. 05). The inhibition of the growth of NB4 cells was not obvious in the combination group than in the solo- medication group(P〉0. 05).Conclusions: The inhibition of the growth of HL60,EOL- 1 cells was more obvious in the combination group of PD0166285 and Doxorubicin than in the solo- medication group.
