不同剂量维生素C对奥沙利铂抗肿瘤效果的影响

THE INFLUENCE OF DIFFERENT DOSES OF VITAMIN C ON ANTITUMOR EFFECT OF OXALIPLATIN:AN ANIMAL EXPERIMENT

目的探讨不同剂量的维生素C(VC)在人胃癌裸鼠模型中对奥沙利铂(L-OHP)抗肿瘤作用的影响。方法建立人胃癌裸鼠皮下移植瘤模型,随机分为8组,每组10只。A组腹腔注射50 g/L葡萄糖注射液,B组腹腔注射L-OHP 6 mg/kg,C、D、E组分别腹腔注射100、200、400 mg/kg的VC,F、G、H组分别腹腔注射100、200、400mg/kg的VC及6 mg/kg的L-OHP,葡萄糖注射液、VC每天给药1次,L-OHP每3 d给药1次,连续用药15 d。观察裸鼠生活状态,测血常规和肝、肾功能;测裸鼠瘤体积,绘制移植瘤生长曲线;处死动物,称取瘤质量,计算抑瘤率,行肿瘤、肝脏病理组织学检查。结果 VC与L-OHP之间存在拮抗性交互作用(F=5.072,P<0.01),VC的浓度越高,对L-OHP抑制肿瘤生长的拮抗作用越强。C、D、E组未表现出肿瘤抑制作用;B、F、G、H组肿瘤生长受到抑制,且随VC浓度增高,抑瘤率下降。结论 VC对L-OHP化疗疗效有拮抗作用,且呈剂量依赖性。

Objective To investigate the influence of different doses of Vitamin C（ VC） on antitumor effect of Oxaliplatin（ LOHP） on a nude mouse model of human gastric carcinoma（ HGC）. Methods A model of subcutaneously transplanted HGC was created in nude mice,which were randomized to eight groups with 10 mice in each group. The mice in group A received 50 g / L glucose; those in group B received L-OHP 6 mg / kg; those in groups C,D,and E received 100,200 and 400 mg / kg of VC,respectively; those in groups F,G and H received 100,200 and 400 mg / kg of VC plus 6 mg / kg of L-OHP. All were given by intraperitoneal injection. For glucose and VC,once a day,and for L-OHP,once every 3 days,lasting for 15 days. Living state of the nude mice was observed,their blood routine,liver and renal functions were tested. Tumor volume was measured. A growth curve of transplanted tumor was drawn. The mice were then sacrificed,weight of tumor was measured and tumor inhibitory rate calcula-ted. The tumors and livers were sent for histology. Results There was an antagonistic interaction between VC and L-OHP（ F = 5. 072,P 〈 0. 01）,the higher the concentration of VC,the stronger the antagonistic effect of L-OHP on inhibiting the growth of tumor. No inhibition of tumor was observed in groups C,D and E. The growth of tumor was inhibited in groups B,F,G and H,the tumor inhibition rate declined along with the increase of the concentration of VC. Conclusion Vitamin C has antagonistic effect on efficacy of Oxaliplatin chemotherapy with dose-dependent manner.
